摘  要:目的 探讨表阿霉素(EPI)、卡介苗(BCG)联合温敏凝胶用于膀胱癌灌注治疗的效果及可行性.方法 将大鼠依据是否给予BCG及EPI进行分组,建立大鼠膀胱癌模型,测定治疗前后大鼠体质量、膀胱重量水平以及尿液中肿瘤坏死因子相关诱导凋亡配体(TRAIL)的变化,并检测细胞增殖抗原标志物(Ki-67)的表达水平.结果 BCG-EPI组膀胱重量[(101.2±8.2)g]明显低于对照组[(124.7±13.3)g,P<0.01];10周末,TRAIL浓度EPI-BCG组为(404.24±71.33) ng/L,较对照组[(242.11±77.24) ng/L]明显升高(P<0.05);EPI-BCG组Ki-67增殖指数为(6.3±2.8)%,较对照组[(13.1±6.9)%]明显降低(P<0.01).结论 BCG、EPI联合温敏凝胶增强了膀胱组织的免疫应答,治疗效果更加显著和持久,减少了频繁膀胱灌注,为临床上治疗膀胱癌提供了新的思路.Objective To study the effectiveness of Epirubicin (EPI) and Bacillus CalmetteeGuerin (BCG) blend a magnetic chitosan hydrogel in the treatment of bladder cancer.Methods Group the rats by whether EPI or BCG was used (each group contains 10 rats),and establish the rats tumor model,then the effect of the treatment was evaluated by measuring rats' body and bladder weight,TNF related apoptosis inducing ligand (TRAIL) concentration in urine and Ki-67 expressive level in bladder tissue.Results Compared with control group,the bladder weight of EPI-BCG group [(101.2 ± 8.2) g] was lighter than the bladder weight of control group [(124.7 ± 13.3) g,P 〈0.01] ; TRAIL concentrations of EPI-BCG group [(404.24 ± 71.33) ng/L] rised significantly compare with control group [(242.11 ± 77.24) ng/L,P 〈 0.05] ; proliferation index of Ki-67 protein of EPI-BCG group [(6.3 ± 2.8) %] was lower than control group [(13.1 ± 6.9) %,P 〈 0.01].Conclusion The magnetic injectable hydrogels combined with both EPI and BCG possesses stronger anti-tumor capacity,which could form a basis for a novel bladder tumor treatment.
