摘  要:采用滤纸片和牛津杯法对分离自大连海参养殖圈海水及海底污泥的506株细菌进行了抗菌活性筛选,获得了1株抗菌活性较强的菌株HS-A465。经形态观察及生理生化实验,结合16S rDNA序列分析,确定为海洋侧孢短芽孢杆菌(Brevibacillus laterosporus)。采用薄层层析和茚三酮显色检测,在其发酵上清液中存在2种具有较强抗菌活性的代谢物质,对每一种成分进行了分离纯化和抑菌实验。结果发现,成分2和3具有显著的抑菌作用。进一步分析表明,成分2和3还具有很强的热稳定性和pH稳定性,对有机溶剂耐受性较好。根据茚三酮显色结果,可以初步推断该活性物质结构上含有游离的氨基酸,初步推断它们是直链的肽类物质。这一研究成果将为海洋抗菌活性物质的进一步开发利用提供理论参考。By using method of antimicrobial testing, a marine HS-A465 was isolated from sea cucumber breeding seawater and sea sludge near Dalian area and was identified as Brevibacillus laterosporus based on its morphological, physiological and characteristics, 16S rDNA identifiable and phylogenetie analysis. Its extract showed obvious antimierobial activity using the filtering paper disk and Oxford cup agar diffusion method. By using TLC and ninhydrin, two components were detected from the liquid culture of Brevibacillus laterosporus HS-A465 strain. The separation, purification and antimicrobial testing of each component were conducted. The results indicated that the component 2 and 3 had significant antimierobial effects. Further study showed that the component 2 and 3 also had thermal stability and pH stability to organic solvents. According to ninhydrin color result, it was speculated that the active substances might be linear chain peptides containing an o^-amino group. This research result would provide theoretical reference for further development and utilization of marine antimicrobial active substances.
