摘 要:目的:采用大孔树脂法分离纯化宣木瓜皂苷。方法:通过比较6种大孔树脂对宣木瓜皂苷的吸附量、解吸率,选出AB-8、D101和XAD-4三种树脂研究吸附特性,确定纯化宣木瓜皂苷效果最好的大孔树脂并优化纯化条件。操作条件:1.25mg/m L宣木瓜皂苷溶液上柱,流速3.0m L/min,上样量45mg;55%乙醇洗脱,流速2.0m L/min。结果:XAD-4树脂对宣木瓜皂苷的最大吸附量为13.64mg/g,吸附时间为2.64h。经XAD-4树脂固定床纯化仅一次后宣木瓜皂苷纯度由10.01%提高到72.41%,回收率74.12%。结论:XAD-4树脂固定床适合用于宣木瓜皂苷的分离纯化,吸附率和解吸率较高,再生性能好。Object :To separate and purify saponins from Chaenomeles speciosa using macroporous resin.Methods= Six different macroporous adsorption resins were evaluated for the static adsorption and desorption capacity for saponins,the better resins were AB-8, D101 and XAD-4.The adsorption behavior of saponins on AB-8, D101 and XAD-4 resins were further investigated to determinate the macroporous resin used to purify saponins and select the suitable conditions.Experimental conditions of the fixed bed were determined as follows:saponins concentration was 1.25mg/mL,flow rate was 3.0mL/min,the amount of sample was 45mg.The elution solvent was .55% ethanol ,flow rate was 2.0mL/min. Results: The results showed that the maximum adsorption capacity of XAD-4 was 13.64mg/g, adsorption time was 2.64h.Through only one cycle of treatment,the content of saponins was raised from 10.01% to 72.41% with an overall recovery of 74.12%.Conclusions:XAD-4 resin fixed bed is suitable to be used to separate and purify saponins for its higher adsorption and desorption rate and its excellent regenerability.
