摘  要:目的:探讨转化生长因子-β1(TGF-β1)标签单核苷酸多态性(SNP)与云南西双版纳地区傣族、哈尼族人群 HBV 感染的相关性。方法采集傣族、哈尼族人群血样共600份,其中每个民族包含健康对照100名,HBV 感染患者200例(包括100例 HBV 自限性恢复患者和100例 CHB 患者),运用基质辅助激光解吸电离飞行时间质谱(MALDITOF-MS)对 TGF-β1基因4个标签 SNP(rs2241715、rs2241716、rs747857、rs8105161)进行基因分型,并构建单体型。结果哈尼族和傣族人群 GGCC 单体型频率分别为0.024和0.000,差异有统计学意义(χ2=4.875,P =0.027)。在傣族和哈尼族人群中, CHB 组 rs2241716位点 G 等位基因显著高于自限性恢复组(傣族:CHB 组 G、A 频率为0.805、0.195,自限性恢复组为0.700、0.300,χ2=5.920,P =0.015;哈尼族:CHB 组 G、A 频率为0.775、0.225,自限性恢复组为0.675、0.325,χ2=5.016,P =0.025)。在傣族人群中,CHB 组 rs2241715位点 GG 基因型和 G等位基因分布频率显著低于自限性恢复组(CHB 组 TT、TG、GG 频率为0.480、0.430、0.090,自限性恢复组为0.400、0.380、0.220,χ2=6.488,P =0.039;CHB 组 T、G 频率为0.695、0.305,自限性恢复组为0.590、0.410,χ2=4.800,P =0.029)。rs2241716位点在 G 隐性模式下(GG/GA+AA),CHB 组与自限性恢复组差异有统计学意义(傣族:P =0.039,OR=1.851,95%CI 为1.033~3.316;哈尼族:P =0.041, OR=1.859,95%CI 为1.024~3.373)。rs2241715位点在 G 隐性模式下(GG/TG+TT),CHB 组与自限性恢复组差异有统计学意义(P =0.013,OR =0.341,95%CI 为0.146~0.796)。GGCC 单体型在HBV 感染组的分布频率显著高于健康对照组(傣族:χ2=4.389,P =0.036;哈尼族:χ2=3.867,P =0.049)。结论TGF-β1基因 GGCC �Objective To investigate the association between tagging single nucleotide polymorphisms (SNP)in transforming growth factor (TGF)β1 gene and hepatitis B virus (HBV) infection in Dai and Hani minorities in Xishuangbanna District, Yunnan Province. Methods The peripheral blood samples of six hundred subjects were collected from Dai minorities and Hani minorities, including 100 healthy controls and 200 HBV-infected individuals (including 100 spontaneous recovery subjects and 100 chronic hepatitis B [CHB]patients)in each minority group.Matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDITOF-MS)was used to genotype the four tagging SNP (rs2241715 ,rs2241716,rs747857,rs8105161)in TGF-β1 gene and construct haplotypes of them.Results The frequencies of GGCC haplotype in Hani and Dai minorities were 0.024 and 0.000, respectively,whose difference has statistical significance (χ2 = 4.875 ,P = 0.027 ).In Hani and Dai minorities,the frequency of rs2241716 G allele in CHB group was significantly higher than spontaneous recovery group (Dai:frequencies of G and A in CHB group were 0.805 and 0.195 ,respectively;whereas those in spontaneous recovery group were 0.700 and 0.300,respectively;χ2 =5 .920,P =0.015 .Hani:frequencies of G and A in CHB group were 0.775 and 0.225 ,respectively;whereas those in spontaneous recovery group were 0.675 and 0.325 ,respectively;χ2 =5 .016,P =0.025).In Dai minorities,frequency of GG genotype at rs2241715 and G allele distribution in CHB group were significantly lower than spontaneous recovery group (frequencies of TT,TG and GG in CHB group were 0.480,0.430 and 0.090, respectively; while those in spontaneous recovery group were 0.400,0.380 and 0.220, respectively;χ2 = 6.488,P = 0.039.frequencies of T and G in CHB group were 0.695 and 0.305 , respectively;while those in spontaneous recovery group were 0.590 and 0.410,respectively;χ2 =4.800, P =0.029).It was significantly different between CHB group and spontaneous recovery g
