摘 要:目的探讨活化巨噬细胞对肝癌细胞侵袭能力的影响及可能机制。方法将人单核/巨噬细胞株THP-1诱导成活化巨噬细胞,并与肝癌细胞株SMMC7721进行非接触式共培养。以Transwell侵袭模型观察活化的巨噬细胞对SMMC7721细胞侵袭能力的影响。再采用ELISA方法检测共培养体系中金属基质蛋白酶9的表达水平。结果经过共培养的肝癌细胞较单独培养的肝癌细胞侵袭力加强,其金属基质蛋白酶9的表达也相应升高。结论活化的巨噬细胞能促进肝癌细胞的侵袭能力,可能与其上调后者表达金属基质蛋白酶有关。Objective To explore the effects of macrophages on invasiveness of hepatocellular carcinoma cell strain and the possible mechanism. Methods Human monocyte/macrophage cell strain THP-1 was induced into activated macrophage, and noncontact co-culturing was made with hepatocellular carcinoma cell strain SMMC7721. The effects of activated macrophages on invasiveness of SMMC7721 cells were observed through a Transwell invasive model. And then, the method of ELISA was used to examine the expression levels of matrix metalloproteinase 9 in the co-culturing system. Results Co-cultured hepatocellular carcinoma cells had stronger invasiveness than simply cultured hepatocellular carcinoma cells, and the expression level of matrix metalloproteinase 9 also increased. Conclusion The activated macrophages can improve the invasiveness of hepatocellular carcinoma cells,and this may be related to the improvement of the expression level of matrix metalloproteinase 9 of the latter.
