胃癌细胞中CD44v6与生存素蛋白的相关性分析

摘　　要：目的探讨胃癌细胞MGC803中生存素蛋白活性降低对CD44v6蛋白表达量的影响。方法将传代后的胃癌MGC803细胞分为实验A、B组和对照组,两个实验组加入不同质量浓度的生存素抗体,对照组加入等量的生理盐水。采用免疫组织化学、蛋白印迹(Western Blot)法检测MGC803细胞样本中CD44v6蛋白表达量的变化。结果与对照组比较,不同质量浓度的生存素抗体均能降低CD44v6蛋白表达;实验B组与实验A组比较降低更明显(P<0.01)。结论抑制生存素蛋白活性可降低胃癌细胞MGC803中CD44v6蛋白的表达。Objective To study the effect of reducing the activities of survivin protein in gastric carcinoma MGCS03 cells to the expression of CD44v6 protein. Methods The cultured gastric carcinoma MGC803 cells were divided into experimental A, B group and the control group. Experimental groups were added with different concentrations of survivin antibody,while the control group with the same amount of saline solution. The expression of CD44v6 protein of MGCS03 cells was detected by immunohistochemical and Western Bloting method, respectively. Results Compared with control group, different concentrations of survivin antibody could reduce CD44v6 protein expressionCompared with the experimental group A, the experimental group 13 reduced more obviously（ P 〈0.01）. Conclusion The inhibition of survivin protein can reduce the expression of CD44v6 protein in MGC803 cells.
