摘 要:为了研究酶法提取山豆根(Sophora tonkinensis Gapnep)总黄酮的最佳工艺条件,对3种酶(纤维素酶、果胶酶、纤维素酶与果胶酶的复合酶)提取山豆根总黄酮的效果进行了比较,选用纤维素酶为提取剂,以山豆根总黄酮得率为考察指标,分别对4个提取参数(提取液p H、提取时间、提取温度及酶加量)进行单因素试验,然后采用正交试验对提取条件进行优化。结果表明,山豆根总黄酮的最佳提取条件为:酶加量0.5%、提取温度70℃、提取时间1.5 h、提取液p H 4.0,在此条件下,山豆根总黄酮的提取率可达到0.73%。该结果可为开发利用山豆根植物资源提供理论依据。To study the optimal technology of extracting total flavonoids from Sophora tonkinensis gapnep,three kinds of enzymes including cellulose enzyme,pectinase and the compound of two enzymes were used to compare the extraction effect with single factor test and orthogonal design.The extraction parameters included p H,enzymatic time,enzymatic temperature and enzyme amount.The results showed that the yield of total flavone extracted from Sophora tonkinensis gapnep were 0.73%under the optimal conditions of enzymolysis 1.5 hours under 70 ℃ and p H of 4.0 with the enzyme amount of 0.5%.It will provide a reference for developing Sophora tonkinensis Gapnep.
