摘 要:为实现北虫草[Cordyceps militaris(L.Fr.)Link]培养基的高效利用,采用闪式提取技术从北虫草培养基中提取多糖。通过单因素试验考察了闪式提取的液料比、提取时间、转速对提取效果的影响,利用正交试验法,优化了北虫草培养基中多糖提取的工艺。结果表明,提取时间和转速对多糖提取效果有显著影响,最佳提取工艺为液料比35∶1(m L∶g),提取时间12 min,转速8 000 r/min。在该条件下,闪式提取多糖得率为3.52%,略高于传统热水回流提取的得率,且只需在室温下进行,提取时间大大缩短,说明闪式提取是一种快速有效提取北虫草培养基中多糖的方法。In order to effecntively utilize t he Cordyceps militaris(L.Fr.)Link medium,flash extraction was used to extract polysaccharide from C.militaris(L.Fr.)Link medium.The effects of liquid to material ratio,extraction time and rotation rate on the extraction rate were investigated with single factor tests.The extraction conditions were optimized with orthogonal design.Results shouved that both extraction time and rotation rate had significant effects on the extraction of polysaccharide.The optimal extraction condition was:liquid to material ratio of 35∶1(m L∶g),extracting 12 min and centrifuging at 8 000 r/min.Under the optimal conditions,the extraction rate was 3.52%,slightly higher than the traditional hot water refluxing extraction.The extraction process was carried out at room temperature,and the time was greatly reduced.It is indicated that flash extraction can be used as a rapid and effective method to extract polysaccharide from C.militaris(L.Fr.)Link medium.
