枣L-半乳糖-1-磷酸磷酸酶cDNA克隆及生物信息学分析

摘　　要：枣果是抗坏血酸含量最高的果实之一,而L-半乳糖-1-磷酸磷酸酶(L-galactose-1-phosphate phosphatase,GPP)基因是抗坏血酸合成中的一个重要基因,至今未见该基因在枣中的相关报道。本研究根据GenBank中登录的苹果、桃和拟南芥等植物GPP保守序列设计引物,采用RT-PCR法,从‘金丝小枣’(Ziziphus jujuba‘Jinsixiaozao’)叶片中克隆出该基因并对其编码的蛋白进行了生物信息学分析。该基因包含完整的开放阅读框为813bp,编码270个氨基酸残基,预测分子量为29.083kDa,理论pI值为5.28,命名为ZjGPP(登录号KJ739593);进一步的生物信息学分析表明,该序列具有典型的肌醇单磷酸化酶结合区域,属于FIG家族蛋白;其氨基酸与其他植物GPP基因具有较高的同源性。进化树分析表明,该基因与桃、苹果和刺梨等蔷薇科植物的GPP亲缘关系较近。Chinese jujube （Ziziphusjujuba Mill. ） is one of the fruits with high ascorbic acid （AsA）. L-galactose-l-phosphate phosphatase gene is an important gene in the AsA biosyn- thetic pathway, which has not been reported in Chinese jujube. In this paper, an 813 hp open reading frame cDNA encoding GPP was cloned from Chinese jujube （Z. jujuba ＇Jinsixi- aozao＇） by RT-PCR method on the basis of the homologous genes of Malus domestica, Prunus persica and Arabidopsis thaliana, etc. in GenBank. The sequence was named ZjGPP （accession number KJ739593） and encoded 270 amino acids, with molecular weight 29. 083 kDa and theoretical isoelectric point 5.28. Conservation analysis of ZjGPP showed that it be- longed to a plant FIG family with a typical zone of inositol monophosphatase; similarity analy-sis showed that the protein had highly homologous analysis showed that GPP from jujube had a closer ca, Malus domestica and Rosa roxburghii. with other known GPP genes; phylogenic relationship with that from Prunus persi-ca, MaLus domestica and Rosa roxburghii.
