摘 要:为探讨丹参酮ⅡA(Tanshinone IIA,Tan IIA)抗动脉粥样硬化(Atherosclerosis,AS)的影响,用脂多糖(Lipopolysaccharide,LPS)体外刺激人外周血单个核细胞(PBMCs)及人单核细胞系(THP-1),通过ELISA和RT-PCR方法分别检测细胞的促炎性细胞因子(IL-1β、TNFα、IL-6)和粘附分子VCAM-1的表达;同时用丹参酮IIA预先处理上述2种细胞,分析丹参酮IIA对LPS诱导细胞因子及粘附分子表达的影响。结果表明,LPS可明显诱导PBMCs和THP-1细胞IL-1β前体、TNFα、IL-6和VCAM-1的表达,而丹参酮ⅡA可明显抑制LPS诱导上述细胞因子及粘附分子表达,表明丹参酮ⅡA能够明显的抑制单核/巨噬细胞介导的炎性应答反应,为探索丹参酮ⅡA抗AS的机制提供了重要的试验依据。To illustrate the mechanism of Tanshinone Ⅱ A anti-atherosclerosis effect, the hu- man peripheral blood mononuclear cells (PBMCs) and human mononuclearcyte THP-1 were pretreated by Tanshinone Ⅱ A, and then primed by LPS. The proinflammatory cytokines (IL- 1β, TNFα and IL-6) and VCAM-1 adhesion molecule were measured by ELISA and RT-PCR. Results showed that LPS significantly induced the expression of IL-1β, TNFα, IL-6 and VCAM-1 in both PBMCs and THP-1 cells. Tanshinone Ⅱ A significantly inhibited the LPS-in-duced expressions of those cytokines and the adhesion molecule, indicating that Tanshinone Ⅱ A possesses the ability to inhibit monocyte-mediated inflammatory response. This study pro vides the important data for further investigations on Tanshinone Ⅱ A anti-atherosclerosis.
