低密度脂蛋白受体LDLR基因在胃癌中的表达及意义

摘　　要：目的检测低密度脂蛋白受体LDLR在胃癌肿瘤细胞系和组织中基因水平的表达,并探讨其与胃癌的相关性。方法利用逆转录聚合酶链反应(RT-PCR)检测7株胃癌细胞系及其56对胃癌组织和癌旁配对组织中LDLR基因的mRNA水平表达状况。并对所得数据做相关性研究分析。结果 LDLR在7株胃癌细胞系的mRNA水平均有表达且有差异,其中N87、MKN45表达较高。LDLR在56对胃癌和癌旁配对组织中的mRNA水平表达有差异,肿瘤组织表达水平71.4%(40/56),高于癌旁配对组织39.3%(22/56),差异有显著性(P<0.01)。结论 LDLR基因在不同胃癌细胞系和胃癌组织中高表达,且胃癌肿瘤组织表达高于癌旁组织,提示低密度脂蛋白受体LDLR基因表达与胃癌相关。Objective To clarify the expression of LDLR in gastric cancer and analyze the relation between LD- LR and GC. Methods RT -PCR was used to detect the expression of LDLR in 7 cell lines,56 pairs of specimens of GC tissue and matched adjacent tissues. Results RT - PCR data confirmed that differential and high expression of LDLR represented at mRNA level in 7 GC cell lines, especially, the expression of LDLR in N87 and MKN45 were higher than others. Furthermore, the expression level of LDLR was higher in the GC tissues compared with matched adjacent tissues. The data showed that over - expression of LDLR were detected in 71.4% （40/56）of GC tissues compared with 39.3% （22/56）of matched adjacent normal tissues, there showed significant statistical difference （P 〈0.01 ）. Conclusion Positive expression of LDLR may have important biological significance and function in carcinogenesis of gastric cancer.
