摘  要:本试验探究苏姜猪心脏脂肪酸结合蛋白质(H-FABP)基因的多态性及其与肉质性状的相关性,采用2种内切酶HinfⅠ和HaeⅢ的PCR-RFLP分子标记技术,分析了40头苏姜猪H-FABP基因5'-上游区和第2内含子区的遗传变异情况。结果显示,试验猪群5'-上游区HinfⅠ酶切位点存在多态性,酶切得到HH、Hh、hh 3种基因型,H等位基因频率为0.65,表现为中度多态(PIC=0.351 5),第2内含子区HaeⅢ酶切位点存在多态性,酶切得到DD、Dd、dd 3种基因型,D等位基因频率为0.70,表现为中度多态(PIC=0.331 8);苏姜猪群的HinfⅠ位点等位基因频率和基因型频率都处于哈迪温伯格平衡状态(P>0.05);H-FABP基因对肌内脂肪含量的影响表现为HH基因型﹥Hh基因型﹥hh基因型及dd基因型﹥Dd基因型﹥DD基因型(P<0.05),对大理石纹的影响表现为HH基因型﹥Hh基因型﹥hh基因型及Dd基因型﹥dd基因型﹥DD基因型(P<0.05)。表明苏姜猪H-FABP基因5'-上游区和第2内含子区的多态性对其肉质性状有重要的影响。Polymorphism of heart fatty acid-binding protein(H-FABP) gene in Sujiang swine and its correlation with meat quality were studied.Polymerase chain reaction-restriction fragment length polymorphysim(PCR-RFLP) with two kinds of restriction enzymes of Hinf I and Hae Ⅲ and molecular marker techniques were applied to analyze the genetic variations of H-FABP gene 5'-upstream region and second intron in 40 Sujiang swine.There was polymorphism in locus of the Hinf I in 5'-upstream region with three genotypes of HH,Hh and hh,and the frequency of allele H was 0.65,showing moderate polymorphism(PIC = 0.351 5).In the second intron,polymorphism was detected in polymorphic locus Hae Ⅲwith three genotypes of DD,Dd and dd,the frequency of allele D was 0.70,indicating moderate polymorphism(PIC= 0.331 8) as well.Both Hinf I gene frequencies and genotype frequencies of tested pigs stayed in the Hardy-Weinberg equilibrium(P〉0.05).H-FABP gene significantly affected the intramuscular fat(IMF) content and marbling content(P〈0.05),following the significance order of HH〉 Hh 〉hh and dd〉Dd 〉DD for IMF,HH〉 Hh〉 hh and Dd 〉dd〉 DD for marbling content.The results suggested that the polymorphisms in 5'-upstream region and second intron of HFABP gene had significant effects on meat quality in Sujiang swine.
