摘  要:目的观察果王素对肺腺癌小鼠移植瘤肺转移的抑制作用,并初步探讨相关机制。方法 40只接种Lewis肺腺癌的C57BL/6J小鼠按随机数字表法随机分成5组,分别为对照组、高剂量果王素组、化疗组(GP方案)、低剂量果王素联合化疗组及高剂量果王素联合化疗组,给予相应药物干预,于接种后20天处死各组小鼠,观察肿瘤肺转移情况,免疫组织化学检测肿瘤T-钙黏蛋白(T-cadherin,T-cad)、乙酰肝素酶蛋白(heparanase,Hpa)的表达,RT-PCR检测肿瘤T-cad mRNA、Hpa mRNA的表达。结果各用药组肺转移结节数明显低于对照组(均P<0.05),高剂量果王素联合化疗组转移结节数明显低于其他组(均P<0.01),高剂量果王素组和高剂量果王素联合化疗组转移抑制率与其他各组比较明显升高,差异均有统计学意义(均P<0.01);对照组T-cad蛋白的表达水平最低,低剂量果王素联合化疗组、高剂量果王素组、高剂量果王素联合化疗组同对照组比较T-cad表达明显增加,差异均有统计学意义(均P<0.01),T-cad mRNA表达与T-cad蛋白表达呈一致性;对照组Hpa的表达水平最高,低剂量果王素联合化疗组、高剂量果王素组、高剂量果王素联合化疗组同对照组比较Hpa表达明显减少,差异均有统计学意义(均P<0.01),Hpa mRNA表达与Hpa蛋白表达呈一致性。结论果王素能明显抑制肺腺癌小鼠移植瘤肺转移,其机制可能与T-cad蛋白及mRNA的表达上调和Hpa蛋白及mRNA的表达下调有关。Objective To investigate the effect of Kiwi essence on the metastasis of lung adenocarcinoma in mice and its mechanism.Methods Forty C57 BL/6J mice bearing Lewis lung adenocarcinoma were randomized into 5 groups:control group,high-dose Kiwi essence group,GP chemotherapy group,low-dose Kiwi essence plus chemotherapy group and high-dose Kiwi essence plus chemotherapy group.Appropriate drug intervention was given to mice in each group.Twenty days after inoculation,the mice were sacrificed,lung metastasis was observed;the protein and mRNA expression of Tcadherin and heparanase in tumor was detected by immunohistochemistry and RT-PCR,respectively.Results The metastatic nodules of lung in treatment groups were significantly less than those in control group(all P 〈 0.05);metastatic nodules were significantly less in high-dose Kiwi essence plus chemotherapy group than those in other groups(all P 〈0.01).The suppressing rate of metastasis was significantly increased in high-dose Kiwi essence group and high-dose Kiwi essence plus chemotherapy group compared to other groups(all P 〈 0.01).Compared to control group,the expression of Tcadherin protein and mRNA was significantly increased in low-dose Kiwi essence plus chemotherapy group,high-dose Kiwi essence group and high-dose Kiwi essence plus chemotherapy group(all P 〈 0.01).Compared to control group,the expression of heparanase protein and mRNA was significantly reduced in low-dose Kiwi essence plus chemotherapy group,high-dose Kiwi essence group and high-dose Kiwi essence plus chemotherapy group(all P 〈 0.01).Conclusion Kiwi essence can significantly inhibit the metastasis of lung adenocarcinoma in mice,which may be associated with the upregulation of T-cadherin expression and the down-regulation of heparanase expression.
