摘  要:目的研究总三帖酸对高糖诱导的星形胶质细胞凋亡的干预作用及机制。方法采用体外分离乳鼠星形胶质细胞培养并分为对照组、模型组、总三帖酸干预组。高糖温孵后提取细胞mRNA和蛋白质分析细胞凋亡蛋白Bax/Bcl2及NADPH氧化酶亚基p22的表达。结果模型组细胞Bax表达上调而Bcl2表达下调,同时p22表达上调。总三萜酸干预后上述蛋白表达部分得到恢复。结论总三帖酸可通过抑制p22上调抑制高糖引起的星形胶质细胞凋亡,减轻细胞的损伤。Objective To investigate the effects of polygala triterpenoid acid on the astrocytes apoptosis caused by high glucose.Methods Cultured astrocytes of neonatol rats were divided into control group,high glucose group and polygala triterpenoid acid group.The expression level of p22 phox,Bax and Bcl2 of three groups were compared.Results The expression levels of p22 phox and Bax were significantly up-regulated and the expression of Bcl2 was down-regulated in high glucose group compared with these of control group.Polygala triterpenoid acid could make the above proteins expression restoration.Conclusion Polygala triterpenoid acid can alleviate the astrocytes damage and apoptosis caused by high glucose by down-regulating the expression level of p22 phox.
