摘  要:目的探讨胃癌中微小RNA(miR)-145的作用及其对c-myc的调控作用。方法取2012年5月—2013年3月在上海交通大学附属同仁医院就诊的51例胃癌患者的胃癌组织和癌旁组织。采用反转录-聚合酶链反应检测miR-145和c-myc表达,并分析其与胃癌临床病理特征的关系。采用Western印迹法检测胃癌细胞株(MKN45、SGC7901)和正常胃黏膜细胞GES-1中miR-145和c-myc的表达。将SGC7901分别转染阴性对照(NC)和miR-145mimics,采用Western印迹法检测c-myc表达的变化。结果胃癌组织中miR-145的表达水平显著低于癌旁组织(P=0.008),c-myc的表达水平显著高于癌旁组织(P=0.000)。不同性别或年龄、有无远处转移的胃癌患者的miR-145和c-myc表达水平的差异均无统计学意义(P值均>0.05);肿瘤浸润深度T2+T3+T4的胃癌患者的c-myc表达水平显著高于肿瘤浸润深度T1的患者(P=0.047),有淋巴结转移(N1+N2+N3)的胃癌患者的c-myc表达水平显著高于无淋巴结转移(N0)的患者(P=0.042),但不同肿瘤浸润深度、有无淋巴结转移的胃癌患者的miR-145表达水平的差异均无统计学意义(P值均>0.05)。人胃癌细胞株MKN45和SGC7901的miR-145表达水平均显著低于胃正常黏膜细胞株GES-1(P=0.001、0.024),c-myc表达水平均显著高于胃正常黏膜细胞株GES-1(P=0.028、0.002)。SGC7901细胞转染miR-145后的c-myc的表达水平显著低于转染NC后(P=0.005)。结论 miR-145作为抑癌基因可以调控胃癌中c-myc的表达。Objective To investigate the role of micro RNA 145 (miR-145) in gastric cancer and its effect on c-myc. Methods Gastric cancer tissues and paracancerous gastric tissues were collected from 51 patients with gastric cancer treated between May 2012 and March 2013 in Shanghai Tongren Hospital. The expression of miR-145 and c-myc were examined by real-time polymerase chain reaction (RT-PCR) and their relationship with clinicopathologic features of gastric cancer was analyzed. The protein expression in gastric cancer cell lines (MKN45, SGC7901) and normal gastric mucosa (GES-1) were detected by Western blotting analysis. After miR- 145 mimics and negative miRNA expression were transfected into SGC7901, the expression of c-myc was detected by Western blotting analysis. Results The expression of miR-145 was significantly decreased and the expression of c-myc was increased in gastric cancer tissue compared to that in paracancerous tissue ( P = 0. 008, P = 0. 000). The expression of miR-145 and c-myc were not correlated to gender and age (all P〉0.05). The expression of cmyc was correlated to the depth of tumor invasion and lymphatic metastasis (P = 0. 047, P = 0. 042), but no significant correlation was found between the expression of miR-145 and the depth of tumor invasion, lymph node metastasis or distant metastasis (all P〉0.05). The expression of miR-145 was significantly decreased in gastric cancer cell lines ( P = 0.001 in MKN45, P = 0. 024 in SGC7901) and the expression of c-myc was increased ( P = 0. 028 in MKN45, P = 0. 002 in SGC7901 ) compared to that in normal gastric mucosa GES-1. The expression of cmyc was down-regulated after transfection compared to negative control (P = 0. 005). Conclusion The tumor suppressor gene miR-145 can regulate the expression of c-myc in gastric cancer.
