摘 要:【目的】通过对血管生成素受体Tie2在血管瘤组织的表达及其甲基化状态的测定,初步探讨其与婴幼儿血管瘤发生、发展的关系。【方法】收集60例血管瘤患儿手术切除的血管瘤标本(其中增生期和消退期标本各30例)及30例正常皮肤组织(包茎患儿包皮环切术切除的包皮组织)。应用甲基化特异性PCR(methylation-specfic PCR,MSP)测定Tie2基因甲基化状态;应用q PCR、Western blotting检测Tie2 m RNA和蛋白表达水平。【结果】30例增生期血管瘤组织、30例消退期血管瘤组织及30例对照组织中Tie2基因启动子区Cp G岛发生甲基化的例数分别为25、14、0例。3组Tie2基因甲基化发生率分别为84.70%、46.64%、0。增生期血管瘤组织甲基化发生率显著高于消退期血管瘤和对照组,差异有显著统计学意义(P<0.01),消退期血管瘤组织甲基化发生率显著高于对照组,差异有显著统计学意义(P<0.01);在增生期和消退期瘤体中Tie2 m RNA和蛋白的表达量明显高于正常包皮组织(P<0.05),其表达在增生期和消退期瘤体之间没有统计学差异(P>0.05)。【结论】婴幼儿血管瘤中存在Tie2基因异常甲基化,且其表达升高,可能参与了血管瘤的发生与发展机制。【Objective】To study the relationship of Tie2 gene with genesis, progress by detecting the expression and methylation in infantile hemangioma.【Methods】60 specimens of infantile hemangioma(30 specimens of proliferating hemangioma and 30 specimens of involuting hemangioma) and 30 specimens of normal foreskins which acquired from 30 boys(undergoing circumcision) were used in this experiment. Methylation of Tie2 gene was examined by using methylation specific PCR(MSP) analysis. The expression of Tie2 receptors m RNA and protein were examined by using q PCR and Western blot analysis.【Results】The positivity rate of promoter methylation of Tie2 gene was significantly higher in proliferating hemangioma than that in the involuting hemangioma and normal foreskins(84.70% VS 46.64% and 0,P〈0.01). Real-time PCR and Western blotting showed that the level of Tie2 m RNA and protein in proliferating and involuting hemangioma was higher than that of the normal foreskins(P〈0.05). There was no statistical difference between proliferating and involuting hemangioma in Tie2(P〉 0.05).【Conclusions】Abnormal expression of Tie2 and methylation of Tie2 gene are found in infantile hemangiomas, which possibly participate in the generation and development of hemangioma.
