摘  要:以梅花鹿鹿角盘为原料,运用响应曲面法优化鹿角盘胶原蛋白提取工艺。以胶原蛋白提取率为指标,在单因素的基础上,选取p H值、加酶量、酶解温度、酶解时间4个因素进行4因素4水平的Box-Behnken试验设计,采用响应曲面法分析4个因素对胶原蛋白提取率的影响,得出鹿角盘胶原蛋白的最佳提取工艺,根据实际情况修正为p H 1.8、加酶量4%、酶解温度37℃、酶解时间4 h。在此条件下,鹿角盘胶原蛋白提取率可达到80.10%。With the antler base of Cervus nippon,extracting technology of collagen was optimized by the response surface methodology. Based on single factor experiments,a four-factor and four-level Box-Behnken design coupled with response surface analysis was carried out to investigate the effects of p H value,enzyme loading,hydrolysis temperature,hydrolysis time on collagen output.The importance of four factors to the extraction of collagen was descended from hydrolysis temperature,hydrolysis time,enzyme loading to p H value. The optimal extracting methods for collagen with antler base was modified as p H 1. 8,enzyme loading 4%,hydrolysis temperature 37℃ and hydrolysis time 4 h. Under the optimized conditions,collagen yield with antler base was 80. 10%.
