摘 要:目的观察胶质细胞源性神经营养因子(GDNF)在小鼠肾脏发育过程中的时空性表达及定位,探讨其与肾发育的关系。方法选取胚龄E12、14、16、18 d胎鼠和生后N1、7、14、21和40 d仔鼠肾脏,应用免疫组织化学技术和免疫荧光技术对小鼠肾组织中GDNF表达进行定位观察;应用蛋白印迹技术对小鼠肾组织中GDNF表达进行定量检测。结果免疫组化和免疫荧光结果:E12 d,GDNF在输尿管芽及生后肾组织微弱表达。随后,在肾小体发育各阶段包括小泡体、逗号小体、S小体、毛细血管袢期肾小体及未成熟期肾小体均有表达,其中,在逗号小体和S小体阶段表达较为强烈;在成熟肾小体中未见表达。随着早期髓质的出现,GDNF在近端小管、远端小管和集合管均有明显的表达。在成熟肾脏中,GDNF主要定位表达于近端小管、远端小管和集合管。蛋白印迹结果:出生前,随着胚龄的增加,GDNF在肾脏表达逐渐增加,在E18 d表达量最高;出生后,随着日龄的增加,GDNF在肾脏表达逐渐减少,在N40 d成鼠肾脏表达量最低。结论肾发育过程中,GDNF对肾小体的形成及肾小管和集合管的分化和功能的维持起关键作用。【Objective】To observe the temporal and spatial expressions of glial cell line-derived neurotrophic factor(GDNF) during kidney development of mouse, and investigate the relationship between GDNF and kidney development.【Methods】The expressions of GDNF were examined by immunohistochemistry and immunofluorescence.The protein content of GDNF in embryonic(including fetal age of 12, 14, 16 and 18 d) and neonatal(including the age of 1, 7, 14, 21 and 40 d) mouse kidneys were measured by Western blot. 【Results】Immunohistochemistry and immunofluorescence results showed that GDNF was expressed weakly in ureteric bud and metanephrogenic tissue at E12 d. Then, GDNF were expressed in vesicle bodies, comma-shaped bodies, S-shaped bodies, capillary loop stage and immature renal corpuscles. Expression of GDNF in mature renal corpuscles was not found. GDNF were also expressed in proximal tubule, distal tubule and collecting duct of developmental and mature kidney. Western blot results showed expression of GDNF in kidney was increased along with embryonic age and reached to the peak at E18 d. Then, expression of GDNF in kidney was decreased along with neonatal age and reached to the lowest at N40 d. 【Conclusions】The expressions of GDNF in developing kidney had temporal and spatial sequence. GDNF may be relative with the development of renal corpuscles and maintain the differentiation and function of proximal tubule, distal tubules and collecting ducts.
