摘 要:观察BK通道在血管平滑肌细胞(vascular smooth muscle cell,VSMC)上的功能及数量发生变化时,对VSMC迁移速度的影响。用脂质体转染法使BK通道在VSMC上过表达,或用BK通道特异性阻断剂IBTX(iberiotoxin)阻断VSMC上BK通道的功能,在这两种情形下,观察VSMC迁移速度的变化。研究结果表明,当过表达BK通道于VSMC,与正常对照相比,其迁移速度明显减慢;反之,当BK通道功能被特异性阻断剂IBTX阻断后,VSMC的迁移速度明显加快。最后认为BK通道的表达水平与VSMC的迁移速度呈负相关,过表达BK通道抑制VSMC的迁移。To investigate the effect of BK channel on migration speed of the vascular smooth muscle cells (VSMC) , when the function and amount of BK channels on VSMC altered. Over-expressed BK channels on VSMC by liposome transfeetion, or blocked the function of BK channels on VSMC by specific blocker iberiotoxin (IBTX) , so as to observe the change of VSMC migration speed in the two conditions. Over-expressed BK channel on VSMC obviously decreased migration speed of VSMC compared with normal control, on the contrary, the migration speed increased when the BK channels were blocked by IBTX. The expression level of BK channel is negatively related with the migration speed of VSMC. Over-expressed BK channel on VSMC inhibited the migration speed.
