摘  要:目的:探讨氢气饱和生理盐水对内毒素(LPS)诱导的小鼠急性肺损伤(ALI)的早期保护作用。方法:将小鼠随机分为正常对照组、氢气饱和生理盐水对照组(H2组)、内毒素损伤组(LPS组)、氢气饱和生理盐水治疗组(LPS+H2组),1或2 h后检测肺湿干重比、肿瘤坏死因子α(TNF-α)和白细胞介素-10(IL-10)m RNA表达量。结果 :与正常对照组相比,LPS组的湿干重比显著增加(P<0.01),TNF-αm RNA表达量持续增加(P<0.01),IL-10 m RNA表达量在2 h时才增加(P<0.05)。与LPS组同一时间相比,LPS+H2组湿干重比降低(P<0.05),TNF-αm RNA表达量先降低(P<0.01),后差异无显著性(P>0.05),IL-10 m RNA表达量差异均无显著性(P>0.05)。结论:LPS诱导的ALI早期,腹腔注射氢气饱和生理盐水能够抑制促炎因子的早期表达,从而减轻内毒素诱导的小鼠急性肺损伤。Objective To evaluate the early protective effect of hydrogen-saturated saline on lipopolysaccharide (LPS) induced acute lung injury (ALI) in mice. Methods Mouse were randomly divided into sham group, the hydrogen-saturated saline control group (H2 group), lipopolysaccharide injury group (LPS group) and hydrogen-saturated saline therapy group (LPS+H2 group). The lung wet/dry weight ratio, tumor necrosis factor alpha (TNF-α) and interleukin-10(IL-10) mRNA expression were measured after 1 hour or 2 hours. Results Compared with sham group, the wet/dry weigh ratio of LPS group significant increased (P 〈 0.01), TNF-α mRNA expression increased (P 〈 0.05), and IL-10 mRNA expression increased(P 〈 0.05) after 2 hours. Compared with LPS group at the same hour, the wet/dry weigh ratio of LPS+H2 group decreased (P 〈 0.05), TNF-α mRNA expression decreased (P 〈 0.01) after 1 hour. But the difierence was not significant alter 2 hours (P 〉 0.05). The difference in IL-10 mRNA expression was not significant (P 〉 0.05). Conclusion In the early stage of ALl induced by LPS, hydrogen-saturated saline could reduce the LPS-induced acute lung injury by inhibiting the early expression of inflammatory cytokines.
