摘 要:目的检测电离辐射后早期SD大鼠肝脏内细胞因子TGF-β1和ɑ-SMA表达的变化,为研究急性放射性肝损伤的发病机制提供实验思路。方法雌性SD大鼠54只随机分为6组,对照组9只、实验1组9只、2组10只、3组8只、4组9只、5组9只(分别为照射后第3、7、14、21、28天)。利用全自动生化分析仪速率法、病理HE染色法、免疫组化Elivision法等技术,建立SD大鼠急性放射性肝损伤模型,观察各组TGF-β1和ɑ-SMA的定位及表达变化。结果各组ALT、AST值均在正常范围,差异无明显统计学意义(P>0.05)。实验组细胞因子TGF-β1和ɑ-SMA分布区域一致,表达于肝脏纤维间隔,中央静脉及汇管区周围细胞的胞浆中,而对照组基本不表达或少量表达。3、4和5组TGF-β1积分光密度值分别为(2 340.58±430.12)、(4 025.67±861.24)和(5 782.31±1 025.61),表达呈进行性增加,差异有统计学意义(P<0.05),5组ɑ-SMA积分光密度值为(2 512.23±504.16),与对照组(240.52±70.22)相比表达明显增加,差异有统计学意义(P<0.05)。结论细胞因子TGF-β1和ɑ-SMA参与了急性放射性肝损伤,其免疫组化测定比血清ALT和AST更早提示放射性肝损伤。[Objective] To detect cytokines TGF-β1 and α-SMA expression changes in SD rat liver in the early post-ionizing radiation, and provide basic data on the pathogenesis of acute radiation-induced liver disease. [ Meth- ods] 55 female SD rats were randomly divided into 6 groups, They are experimental groups 1 to 5 (3, 7, 14, 21, 28 days after irradiation) and the control group. The automatic biochemical analyzer rate method, pathological HE stain- ing, immunohistoehemical Elivision techniques were used to the establish SD rat model of acute radiation-induced liver disease. TGF-β1, α-SMA positioning and expression in each group were observed. [Results] No significant dif- ference of AST and AST were observed among 6 groups (P 〉0.05). The cytokine TGF-β1, α-SMA distribution area of experimental group were consistent expression in the liver fibrous septa, central vein and portal area surruunding cy- toplasm, whereas the control group did not express basic or little expression. TGF-β1 integrated optical density values in group 3, 4, 5 were (2 340.58±430.12), (4 025.67±861.24), (5 782.31±1 025.61), the expression was progressively in- creased, the difference was significant (P 〈0.05). α-SMA integral optical density value in group 5 was (2 512.23± 504.16), and compared with (240.52±70.22) of the control group was significantly increased, the difference was sig- nificant (P 〈0.05). [ Conclusion] Cytokine TGF-β1, α-SMA involved in acute radiation-induced liver disease. Rel- ative to serum ALT, AST testing, their immunohistoehemical earlier prompted radiation-induced liver injury.
