摘 要:线粒体与疾病的发生、发展、治疗密切相关。以线粒体为靶向的药物分子设计研究,已成为药学、化学和生命科学的热门领域。目前,以线粒体为靶目标的抗肿瘤药物机制主要集中在线粒体跨膜电势累积、电子传递链复合物抑制、膜渗透转化孔开启、mt DNA调节、糖酵解通路调节等。该文简述了线粒体靶向药物的研究进展,重点概述了以线粒体为靶向的,基于线粒体膜电势、电子传递链、渗透转化、DNA、代谢等药物的作用机制。Mitochondria play an important role in a lot of physical activities,such as energy production,apoptosis and carcinogenesis.Mitochondrial dysfunctions not only influence on cell growth,metabolism,and proliferation,but also trigger mitochondrial disease and apoptosis.Research on mitochondrion targeting drug designing has been a hot topic in pharmacy,chemistry and life sciences.Recently,research on mitochondrion targeting drug designing focuses on accumulation of mitochondrial transmembrane potential,inhibition of electron transfer chain complexes,opening of mitochondrial permeability transition pore(MPTP),mtDNA regulation,glycolytic pathway regulation,etc.Here is to make a review of the research progress in mitochondrion targeting drugs,especially the mechanisms of such drugs based on mitochondrial membrane potential,electron transfer chain,permeability transition,DNA and metabolism,etc.
