摘 要:将80只SPF级ICR小鼠按单纯随机抽样方法分为正常对照组(等体积生理盐水)、PFOA低剂量组(1 mg·kg-1·d-1)、PFOA中剂量组(5 mg·kg-1·d-1)、PFOA高剂量组(25 mg·kg-1·d-1)、GEN干预对照组(20 mg·kg-1·d-1)、GEN干预PFOA低剂量组(GEN:20 mg·kg-1·d-1、PFOA:1 mg·kg-1·d-1)、GEN干预PFOA中剂量组(GEN:20 mg·kg-1·d-1、PFOA:5 mg·kg-1·d-1)、GEN干预PFOA高剂量(GEN:20 mg·kg-1·d-1、PFOA:25 mg·kg-1·d-1)组,每组10只,采用灌胃方法,通过计算肝脏比体重系数,检测肝组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、乳酸脱氢酶(LDH)、丙二醛(MDA)及一氧化氮(NO)指标的水平,探讨了三羟异黄酮(GEN)对不同剂量的全氟辛酸(PFOA)致小鼠肝脏氧化损伤的保护作用。结果表明:PFOA导致肝脏肿大,增加了小鼠肝脏系数,并随着剂量增加而加重。高剂量的PFOA能明显增高肝脏组织中MDA及NO含量(P<0.05),低、中、高剂量的PFOA均能提高LDH活性和降低SOD、GSH-Px的活性(P<0.05),而GEN在干预PFOA各剂量组中能显著提高GSH-Px的活性和降低LDH活性(P<0.05),在干预PFOA高剂量组中能提高SOD的活性(P<0.05)。GEN对PFOA致肝脏组织的脂质过氧化损伤有一定的拮抗作用。80 mice were distributed randomly into normal control group( the same dose of normal saline),PFOA low dose group( 1 mg·kg^- 1·d^- 1,PFOA medium dose group( 5 mg·kg^- 1·d^- 1,PFOA high dose group( 25 mg·kg^- 1·d^- 1,GEN intervention group( 20 mg·kg^- 1·d^- 1,GEN intervention low dose group( GEN: 20 mg·kg^- 1·d^- 1nd PFOA: 1 mg·kg^- 1·d^- 1,GEN intervention medium dose group( GEN: 20 mg·kg^- 1·d^- 1nd PFOA: 5 mg·kg^- 1·d^- 1,GEN intervention high dose group( GEN: 20 mg·kg^- 1·d^- 1nd PFOA: 25 mg·kg^- 1·d^- 1. The mice were weighed every day and their behavior were observed.After 14 days,the mice were killed and liver indexes were calculated. Meanwhile,the activity of superoxide dismutase( SOD),glutathione peroxidase( GSH-Px),lactate dehydrogenase( LDH) and the content of malondialdehyde( MDA) and nitric oxide( NO) were also assayed. The results showed that the content of MDA and NO were increased dramatically in PFOA high dose group( P 〈0. 05),the activity of LDH in hepatic tissue of PFOA groups were increased dramatically while the activity of SOD and GSH-Px was decreased obviously compared with the control group in 3 dose group of PFOA( P 〈0. 05),GEN could increase the activity of GSH-Px and decrease the activity of LDH in 3 GEN intervention groups( P 〈0. 05),while the activity of SOD was increased in GEN intervention high dose group( P 〈0. 05). Supplementation of GEN during PFOA exposure may have certain protective effect on inhibition caused by PFOA exposure.
