Wistar大鼠曲马多依赖模型建立方法的探讨及体会

摘　　要：目的成功建立曲马多(TR)依赖大鼠动物模型。方法将80只Wistar大鼠平均分成4个实验组(正常对照组20只、空白对照组20只、TR依赖组20只、TR戒断组20只),观察TR腹腔注射给药及停药戒断过程中大鼠躯体依赖与精神依赖的情况。结果与正常对照组及空白对照组比较,TR戒断组和TR依赖组的大鼠体重都有所下降(P<0.01);给药5分钟后,TR依赖组及TR戒断组大鼠对声音及触摸刺激敏感;在条件性位置偏爱实验中,TR依赖组及TR戒断组大鼠表现出偏爱性(P<0.01)。TR戒断组停止给药后,大鼠表现出烦躁和易激惹等现象。结论本实验中大鼠对TR产生了依赖,可以证明成功建立了TR依赖大鼠动物模型。Objective To successfully established tramadol rely on animal model of rats. Methods An average of 80 Wistar rats were divided into four group （ normal control group, blank control group only 20, TR depend on the group of 20, the TR withdrawal only group of 20） , and observed by intraperitoneal injection of tramadol medication and drug withdrawal withdrawal process cuhk rat physical dependence and psychological dependence. Results Compared with normal control group and blank control group, the TR withdrawal group and TR depend on body weight of rats were decreased （ P 〈 0.01 ） , After 5 minutes, TR dependence and TR withdrawal group rats is sensitive to voice and touch stimulation, In the conditioned place preference experiments, TR dependence and TR withdrawal group rats showed a preference for sex （ P 〈 0. 01 ）. TR withdrawal group stopped after dosing, rats showed the phenomenon such as restlessness and irritability. Conclusion This experimental cuhk for tramadol dependence, can prove that successfully established tramadol rely on animal model of rats.
