摘 要:目的探讨蚓激酶微乳及其与片仔癀联用对CCl4诱导的大鼠肝纤维化的作用研究。方法 SD大鼠采用CCl4法诱发大鼠肝纤维化模型,SD大鼠随机分成正常组、阴性组、模型组、蚓激酶微乳组(YR)、片仔癀组(P)、蚓激酶微乳+片仔癀组(YRP),8周末取血测定血浆丙氨酸氨基转移酶(ALT)、天门冬氨酸转氨酶(AST)活性。给药8周后取肝组织进行病理学相关检查以观察大鼠肝组织纤维化程度,同时进行mmp13表达免疫组化检测。结果与模型组相比,YRP、YR、P3组血清转氨酶ALT和AST及其比值AST/ALT均降低,有显著性差异;YRP、P、YR组HE染色显示的肝小叶结构破坏程度、纤维组织增生及炎性细胞浸润程度均较模型组轻,其中YRP组表现更甚;YRP、P、YR组mmp-13免疫组化胞浆棕黄色着色明显增多,提示mmp-13表达皆高于模型组,有显著性差异,YRP组仍强于P组和YR组。结论片仔癀或蚓激酶微乳对改善大鼠肝纤维化程序有一定的抑制作用,片仔癀与蚓激酶微乳联合用药对大鼠肝纤维化有较明显抑制作用。OBJECTIVE Research of combind Oral Lumbrokinase Microemulsion with PienTze Huang effect on CCl4 induced liver fibrosis in rats.METHODS SD rats are using CCl 4 method to induce model rats′liver fibro-sis.SD rats were randomly selected into normal group ,negative group,model group,Lumbrokinase Microemulsion group (YR),PienTze Huang group (P),Lumbrokinase Microemulsion +PienTze Huang group (YRP).Testing ac-tivity of Alanine aminotransferase ( ALT) and Aspartate aminotransferase ( AST) in blood plasma at the end of week 8.Liver tissues were taking after 8 weeks of dosing ,using for pathological examination to observe the effect on liver fi-brosis in rats,at the same time doing mmp13 expression immunohistochemical detection .RESULTS There were significant differences showed:decreased serum transaminase ALT ,AST and the ratio of AST/ALT in all three YRP, YR,P groups compare to the model group;HE staining of all three YRP ,P,YR groups showed:the damage degree of hepatic lobule ,level of fibrous tissue hyperplasia and inflammatory cell infiltration were all less than model group ,es-pecially YRP group showed the least;YRP,P and YR group had significantly increased brown colour in MMP 13 im-munohistochemicalcytoplasm ,which means their mmp 13 expressions are all higher than model group ,there were sig-nigicant differences.The expression of YRP group is still stronger than P group and YR goup .CONCLUSION Pi-enTze Huang or Lumbrokinase microemulsion has some inhibiting effect on liver fibrosis in rats .Combind Lumbroki-nase microemulsion with PienTze Huang can enhance effect and shows obvious inhabitation on liver fibrosis in rats .
