HPLC法同时测定利脑心胶囊中6种成分的含量

摘　　要：目的：建立同时测定利脑心胶囊中丹参素、原儿茶醛、丹酚酸B、芍药苷、阿魏酸和葛根素含量的方法。方法：采用高效液相色谱法。色谱柱为Agilent Zorbax Eclipse XDB-C18,流动相为乙腈-甲醇-0.5%磷酸水溶液（梯度洗脱）,流速为1.0 ml/min,检测波长为280 nm、230 nm、250 nm、320 nm,柱温为30℃,进样量为10μl。结果：丹参素、原儿茶醛、丹酚酸B、芍药苷、阿魏酸、葛根素的质量浓度分别在16.54~264.58、1.44~22.46、187.20~1 497.60、23.71~379.39、0.20~1.56、2.72~42.43μg/ml范围内与各自峰面积积分值呈良好的线性关系（r=0.999 9、0.999 9、0.999 7、0.999 9、0.999 5、0.999 9）;精密度、稳定性、重复性试验的RSD≤1.99%;平均回收率分别为98.37%、98.28%、98.74%、98.15%、98.38%、98.29%,RSD分别为0.63%、0.88%、0.40%、1.31%、2.55%、0.94%（n=6）。结论：该方法操作简单、准确、重复性好、灵敏度高,可用于同时测定利脑心胶囊中6种成分的含量。OBJECTIVE： To establish a method for the content determination of danshensu, protocatechuic aldehyde, salviano- lic acid B, paeoniflorin, ferulic acid and puerarin in Linaoxin capsules. METHODS： HPLC method was adopted. The separation was performed on Agilent Zorbax Eclipse XDB-C,8 column with mobile phase composed of acetonitrile-methanol and 0.5% H3PO4 aqueous solution （gradient elution） at the flow rate of 1.0 ml/min. The detection wavelength was set at 280 nm, 230 nm, 250 nm and 320 rim. The column temperature was set at 30 ℃ , and injection volume was 10 μl. RESULTS： The linear range were 16.54-264.58 μg/ml for danshensu（r=0.999 9）, 1.44-22.46 μg/ml for protocatechuic aldehyde（r=0.999 9）, 187.20-1 497.60 μg/ml for salvianolic acid B（r=0.999 7）,23.71-379.39 μg/ml for paeoniflorin（r=0.999 9）,0.20-1.56 μg/ml for ferulic acid （r=0.999 5）, 2.72-42.43 μg/ml for puerarin （r=0.999 9）, respectively. RSDs of precision, stability and repetition tests were ≤ 1.99%. The aver- age recoveries were 98.37% （RSD=0.63% , n=6） , 98.28% （RSD=0.88% , n=6） , 98.74% （RSD=0.40% , n=6） , 98.15% （RSD=1.31% ,n=6）, 98.38% （RSD=2.55% ,n=6） and 98.29% （RSD=0.94%, n=6）. CONCLUSIONS： The method is simple, accurate, reproducible and sensitive, and can be used for simultaneous determination of 6 components in Linaoxin capsules.
