Survivin、Fhit蛋白在喉癌组织中的表达及临床意义

摘　　要：目的观察喉癌组织中存活素(Survivin)、脆性组氨酸三联体基因(fragile histidine traid,Fhit)的表达,探讨Fhit与Survivin在喉癌组织中的相关性。方法 2006年10月—2007年8月、2008年7月—2013年12月手术治疗并病理证实的92例喉鳞癌标本及相应癌旁组织(距离癌边缘10 mm),25例喉正常黏膜,采用免疫组化链霉卵白素-生物素复合体(SABC法)检测Survivin、Fhit蛋白的表达。结果 Survivin在喉癌组织中的阳性率明显高于癌旁及喉正常黏膜组织(P<0.05),与临床分期有关(P<0.05),与年龄、淋巴结转移、分化程度无关(P>0.05)。Fhit蛋白在喉癌组织中的阳性率明显低于癌旁及喉正常黏膜组织,差异有统计学意义(P<0.05),与临床分期和淋巴结转移有关(P<0.05),与年龄、分化程度无关(P>0.05)。Fhit与Survivin呈负相关(P<0.05)。结论 Survivin蛋白高表达和Fhit蛋白低表达在喉癌的发生、发展上可能有协同作用。联合检测可能成为喉癌新的分子指标。Objective To observe expressions of survivin and fragile histidine triad（ Fhit） protein in laryngocarcinoma tissues and to discuss their relationships. Methods Samples of 92 patients（ group A） with laryngeal squamous cell carcinoma,who were confirmed by pathology after operation during October 2006 and August 2007,and during July2008 and December 2013,and the corresponding ambient carcinoma tissues（ 10 mm from the tumor edge,group B）,and 25 cases（ group C） of normal laryngeal mucosa were detected for survivin and Fhit protein expressions,using immunohistochemical strept avidin biotin complex（ SABC）. Results The survivin positive rate in group A was significantly higher than that in group B and C（ P〈0. 05）. Survivin expression was correlated with clinical stages（ P〈0. 05）,but was not correlated with age,lymphnode metastasis and differentiation degrees（ P〉0. 05）. The positive rate of Fhit protein in group A was significantly lower than that in group B and C,and the differences were statistically significant（ P〉0. 05）. Fhit expression was correlated with clinical stages and lymphnode metastasis（ P〈0. 05）,but was not correlated with age and differentiation degrees（ P〈0. 05）. Fhit was negatively correlated with survivin（ P〈0. 05）. Conclusion Occurrence and development of high expression of survivin protein and low expression of Fhit protein in laryngocarcinoma tissues may have a synergistic effect,therefore combined detection may be a new molecular indicator of laryngocarcinoma.
