摘  要:目的在钛种植体表面制备羟基磷灰石(hydroxyapatite,HA)和神经生长因子(nerve growth factor,NGF)复合涂层,为改善种植体骨感知能力,提高种植体与周围骨组织的结合提供思路。方法将钛种植体浸泡于模拟体液中获得HA钛种植体试件,将NGF与硫酸软骨素(chondroitinsulfate,CS)混合,冷冻干燥后形成NGF-CS纳米颗粒,将纳米颗粒与钛种植体试件共同浸泡于钙磷溶液中,获得HA-NGF钛种植体。种植体NGF的释放量采用酶联免疫吸附剂测定法测定,释放液中NGF活性采用PC12细胞存活法进行测定。结果成功制备出HA-NGF钛种植体试件,且试件有长达20 d的NGF缓慢释放,并且含NGF的试件组PC12细胞生存活性高于不含NGF的试件组,差异具有统计学意义(P<0.05)。结论钛种植体的HA-NGF复合涂层可以缓慢释放具有活性的NGF。Objective To form a complex coating of hydroxyapatite( HA) compound nerve growth factor( NGF) on the titanium( Ti) surface,which provided a thought of modifying the ability of osseointegration and improving the osseointegration. Methods Ti was subjected to soaking in simulated body fluid( SBF) to obtain the sample of HA-Ti. Meanwhile,the nanocomplex was fabricated by using an ionic interaction between NGF and chondroitinsulfate( CS). After that,the nanocomplex was precipitated on the HA-Ti surface,along with the addition of calcium / phosphate solution, the final product was formed as HA-NGF-Ti. The release of NGF was evaluated by ELISA. In addition,the viability of NGF was detected according to survival rate of PC12 cell. Results The sample of HA-NGF-Ti was fabricated successfully. The immobilized NGF was released from the sample HA-NGF-Ti in a moderate rate for 20 days. When PC12 cells were seeded on different Ti substrates,cell proliferation was faster in the HA-NGF-Ti group,as compared to other groups,there was significant difference( P 〈 0. 05). This study demonstrated that the delivery system of NGF was effective on the experiment group. Conclusion The activated NGF can obtain a controlled release from the HA-NGF complex coating on titanium surface.
