摘  要:目的:探讨转化生长因子活酶激酶抑制剂TAK1抑制剂5Z-7-oxozeaenol对大鼠脑缺血再灌注损伤的保护作用。方法:线栓法制备大鼠脑缺血再灌注模型。健康雄性SD大鼠36只(250~320g),随机分为6组:假手术组、缺血组、溶剂组和5Z-7-oxozeaenol低剂量、中剂量和高剂量组。再灌注24h后进行神经功能评分(NBS),TI'C染色测定梗死面积,TUNEL染色检测神经细胞凋亡,并检测大鼠脑组织匀浆中SOD、MDA、TNF-α、IL-1β变化。结果:与缺血组相比,5Z-7-oxozeaenol高剂量组脑梗死容积明显减少,神经行为学评分提高,缺血半暗区神经细胞凋亡数减少(P〈0.05),SOD活性明显升高,MDA、TNF-α、IL-1β含量明显降低(P〈0.05)。结论:转化生长因子活酶激酶抑制剂对大鼠脑缺血再灌注损伤有明显的保护作用,其机制可能与抑制TAK1活化、抑制细胞凋亡有关。Objective:To investigate the protection effeet of 5Z--7--oxozeaenol on the cerebral ischemia-- reperfusion (I/R) injury in rats. Methods:Thirty--six male SD rats weighing 250--320 g were randomly di- vided into 6 groups (n = 6 each) : sham operation group (group Sham), I/R group (group I/R), DMSO group (group Vehicle),low, middle and high dose 5Z--7--oxozeaenol groups. Garcia method was used to score the neurological function and triphenyl tetrazolium chloride (TTC) and computer aided video system were used to measure the infarct volumes. TUNEL(+) cell were counted. At the same time,the levels of SOD,MDA,TNF--a, IL--1β in brain tissue wrere determined. Results: The neurological behavior scores were better in 5Z--7--oxozeaenol (high dose) group than those in control group,the infarct volumes were significantly smaller than those in control group (P〈0.05). Compared with I/R group,5Z--7--oxozeaenol treatment dose--dependently significantly decreased TUNEL(+) cell counts in ischemic penumbra (P〈 0.05). Conclusion: 5Z--7--oxozeaenol attenuates cerebral ischemia and reperfusion injury via depressing of neuronal apoptosis in rats.
