摘  要:目的探讨凋亡抑制基因Livin在喉鳞状细胞癌(laryngeal squamous cell carcinoma,LSCC)组织中的表达及其与p53和Bcl-2表达的关系,探讨Livin在LSCC中的作用机制。方法收集LSCC标本42例,声带息肉组织20例,采用免疫组织化学SP法检测LSCC组织和声带息肉组织中Livin、p53和Bcl-2蛋白的表达。结果 LSCC中Livin表达阳性率为71.4%(30/42),显著高于声带息肉组织(χ2=27.679,P<0.01)。临床分期Ⅲ+Ⅳ期Livin阳性表达率显著高于Ⅰ+Ⅱ期(χ2=7.087,P<0.01);有淋巴结转移者的Livin阳性表达率高于无淋巴结转移者(χ2=5.458,P<0.05)。Livin蛋白的表达与p53蛋白表达呈正相关(r=0.665,P<0.05),与Bcl-2蛋白表达呈正相关(r=0.657,P<0.05)。结论 Livin在LSCC组织中异常表达,有望成为LSCC诊断和基因治疗的新靶点。凋亡相关基因p53与Bcl-2基因的异常表达在喉癌癌变中起协调作用。OBJECTIVE To study the expression of Livin——the inhibiting gene of Apoptosis in laryngeal squamous cell carcinoma(LSCC) as well as its correlation with p53 and Bcl-2, and further explore the mechanism of role of Livin in the development of LSCC. METHODSThe expression of Livin, p53 and Bcl-2 were studied with SP immunohistochemical method in 42 collected samples of LSCC and 20 samples of polyps of vocal cord. RESULTS The positive rate of Livin expression in LSCC(71.4%, 30/42) was much higher than that in polyp of vocal cord(χ 2=27.679, P 〈0.01). The positive rate at clinical Ⅲ+Ⅳ stage patients was much higher than that at clinical Ⅰ+Ⅱ stage patients(χ 2=7.087,P 〈0.01), while the positive rate of Livin in patients with cervical lymph node metastasis was higher than those who without cervical lymph node metastasis(χ2=5.458,P〈0.05). The expression of Livin was positively correlated with p53 expression(r =0.665, P〈0.05) and Bcl-2 expression(r =0.657, P〈0.05). CONCLUSION The abnormal expression of Livin in LSCC can be a new target for diagnosis and treatment of the LSCC. The abnormal expression of p53 and Bcl-2 may play a coordinating role in the development and progression of laryngeal carcinoma.
