摘 要:目的研究脑脉泰影响脑组织神经生长因子(NGF)、胶质纤维酸性蛋白(GFAP)的表达,进而改善慢性脑缺血大鼠的认知功能。方法采用大鼠双侧颈总动脉永久结扎法(2-VO)制备慢性脑缺血模型。术后分别给予脑脉泰不同剂量灌胃治疗6 w。应用Morris水迷宫测定大鼠的学习记忆能力、HE染色观察海马CA1区神经元形态、免疫组化方法检测大鼠脑组织NGF及GFAP的表达。结果与假手术组相比,模型组大鼠的学习能力和记忆能力降低,脑组织中NGF表达减少(P<0.01),GFAP表达增多(P<0.01);脑脉泰给药能够改善大鼠的记忆和学习能力,增加脑组织中NGF表达(与模型组相比,低剂量组P<0.05,高剂量组P<0.01),降低GFAP表达(与模型组相比,低、高剂量均为P<0.01)。结论脑脉泰可能通过提高脑组织NGF和降低GFAP表达来改善慢性脑缺血大鼠的认知功能。Objective To explore the effects of Naomaitai on expression of NGF and GFAP in brain,to elucidate mechenism of Naomaitai on improving the cognitive function following chronic cerebral ischemia in rats. Methods The rat model of chronic cerebral ischemia was established by bilateral general cervical artery ligation( 2-VO). After the operation,different dose of Naomaitai was lavaged to the model rats for six weeks. The learning and memory function of rats were tested by Morris water maze. The change of pyramidal cell in hippocampus CA1 region was observed by HE staining. Using immunohistochemistry to detect the expression of NGF and GFAP in brain. Results The learning and memory function of model rats declined compared with the sham-operated group. The expression of NGF was decreased( P 〈0. 01) and the expression of GFAP was increased( P〈 0. 01). Naomaitai could improve the cognitive function following chronic cerebral ischemia in rats by increasing the expression of NGF( low dose group P〈 0. 05,high dose group P 〈0. 01) and decreasing the expression of GFAP( low dose group and high dose group P 〈0. 01) compared with the model group. Conclusions Naomaitai can improve the cognitive function following chronic cerebral ischemia in rats,it may play a role by increasing the expression of NGF and decreasing the expression of GFAP.
