摘 要:目的探讨根皮苷对血吸虫病肝纤维化小鼠肝组织中转化生长因子β1(TGF-β1)和人血小板衍生生长因子BB(PDGF-BB)表达的影响。方法用腹部贴片法感染昆明小鼠,建立血吸虫病肝纤维化模型,第6周末,肝纤维化模型建立;用吡喹酮杀虫两天后,用低、中、高剂量根皮苷(10、20、40 mg·kg-1·d-1)治疗,同时设病理模型组、空白对照和阳性对照(INF-γ)组;给药8周后,采用免疫组织化学检测各组大鼠肝脏组织中TGF-β1和PDGF-BB的表达,做HE染色测量血吸虫的虫卵结节。结果 TGF-β1和PDGF-BB含量在用药第4周到第8周呈降低趋势,且高剂量根皮苷组的各项指标最接近空白组,抑制血吸虫病肝纤维化作用明显;HE染色显示高剂量根皮苷组的肝脏变性坏死减轻明显,小叶结构完整;高剂量根皮苷组的TGF-β1和PDGF-BB含量与模型组、吡喹酮组、低剂量根皮苷组和中剂量根皮苷组的都有显著性差异(P<0.01);与IFN-γ组比较无显著性差异(P>0.05);根皮苷各剂量组和IFN-γ组的计分较模型组显著降低(P<0.01),根皮苷低、中量组的计分与IFN-γ组的比较有显著性差异(P<0.05),根皮苷各剂量组之间比较有显著性差异(P<0.05)。结果表明:根皮苷剂量呈量效增强关系。结论根皮苷能降低血吸虫病肝纤维化小鼠肝脏中TGF-β1和PDGF-BB的表达,使血吸虫虫卵结节减小,使血吸虫病肝纤维化程度减轻。OBJECTIVE To explore the effects of phlorizin on TGF - β1 and PDGF - BB expression in liver fibrosis mice hepatic tis- sue induced by Schistosomiasis japonicum. METHODS Kunmiug mice were infected by cercariae of S. japonicum by abdominal patch method. At the end of 6th week, liver fibrosis model was made successfully. Experimental establishment of the pathological model group, blank control, and positive control ( INF - gamma) group. The mice were labeled as blank group, Praziquantel group, positive control (interferon gamma)group and low(10 mg·kg^-1 ·d^-1) ,middle(20 mg·kg^-1 ·d^-1) and high(40 mg·kg^-1 ·d^-1) dose phlorizin. After administration eight weeks, the expression of TGF -β1 and PDGF - BB in the liver tissue was detected by immunohistochemical stai- ning. Schistosome eggs nodule size was measured by HE staining. RESULTS The levels of TGF -β1 and PDGF - BB decreased from 4th week to 8th week, and the level of those was closest to blank group in high dose phlorizin group. HE staining solution showed liver degeneration and necrosis were obvious reduced and the lobular structure was the most complete degeneration in high doses of phlorizin group. TGF- β1 and PDGF- BB in high doses phlorizin group contented with the control group, the Praziquantel group, low- dose apigenin group and middle - dose phlorizin group had a significant difference (P 〈 0.01 ). As for IFN - ~/group, there was no significant difference(P 〉 0.05 ). Compared with IFN-7 group, the low, middle and high dose of phlorizin group significantly reduced( P 〈 0.01 ). Compared with IFN - 7 group, the low and middle dose of phlorizin group had significant difference ( P 〈 0. 05 ). The results showed the dose of phloridzin processed dose - effect enhanced relationship. CONCLUSION Phlofizin can reduce the expression of TGF-β1 and PDGF- BB in liver fibrosis mice hepatic tissue induced by schistosomiasis, cut down schistosome eggs nodule size, and mitigated schistosomiasis liver fibrosis.
