青蒿鳖甲汤含药血清对AML-CR患者CD34^+细胞源DC诱导过程中TNF-α的影响

摘　　要：目的观察透毒复方青蒿鳖甲汤对急性残留白血病(AML-CR)患者CD34+细胞源树突状细胞(DC)诱导过程中对TNF-α含量及DC成熟的影响。方法采取AML-CR患者骨髓,体外扩增从中分离的CD34+细胞,以高、中、低剂量青蒿鳖甲汤含药血清联合细胞因子诱导为DC,收集第0、6、9天上清并观察DC形态,ELISA法测TNF-α含量,流式细胞术检测DC表面抗原表达。结果含药血清均能促进DC的成熟,降低TNFα含量,与正常兔血清组、细胞因子组有显著差异(P<0.05),使DC较高表达CD83、CD86、CD1a、HLA-DR。结论青蒿鳖甲汤含药血清联合细胞因子可促进AMLCR患者来源的CD34+细胞诱导成为成熟DC,并降低TNF-α含量。Objective To observe the effect of QingHaoBieJia Decoction on TNF - oL during the differentiation and maturation of CD34^ ＋ cell -derived Dendritic Cells （DC） from the Acute Myelogenous Leukemia in Complete Remission（ AML- CR） patients. Methods Amplify the CD34^ ＋ cells of AML - CR patients in vitro. Culture them with the Medicated Serum of QingHaoBieJia De- coction and Cytokines, and detect the TNF - a from the 0th, 6th, 9th day＇s supernatant serum by ELISA kit. Observe the morphol- ogy of DC during its growing and detect its expression of CD80, CD83, CD86, CDla, HLA- DR by FCM. Results The Medicated Serum combined with cytokines could promote the maturation and expression of CD83, CD86, CD1 a, HLA -DR from DC, and re- duce the secretion of TNFa. There was significant difference between Medicated Serum group and other groups（ P 〈0.05 ）. Con- clusion The Medicated Serum combined with cytokines can promote the maturation of CD34^ ＋ Cell - derived DC, and reduce the secretion of TNF - a.
