摘 要:目的探讨新型免疫分子TIPE2对肝癌细胞转移与侵袭的影响作用。方法取35例肝癌患者外周血单核细胞及40例健康对照样本,采用实时荧光定量法检测外周血PBMCs中TIPE2的表达水平,采用划痕实验和Transwell试验检测TIPE2对BEL7402细胞转移与侵袭的影响。结果肝癌病人PBMCs中TIPE2的表达下调,且其下调程度与肿瘤分期相关,细胞转染实验表明TIPE2上调可以抑制肝癌细胞系的转移与侵袭。结论 TIPE2可能作为一种新型的抑癌基因在肿瘤的发生发展中起重要生物学作用。This study designed to investigate the influence of new immune molecule TIPE2(tumor necrosis factor-alpha-induced protein 8-like 2) on the metastasis and invasion of hepatoma cells. Firstly, we examined the m RNA expression levels of TIPE2 in peripheral blood mononuclear cells(PBMCs) from 35 HCC patients and 40 healthy controls by real-time reverse transcription-polymerase chain reaction analysis(RT-PCR). Then, we examined the impact of TIPE2 on metastasis and invasion of BEL7402 cell by wound scratch assay and Transwell method. The results showed the TIPE2 m RNA expression was significantly decreased in HCC patients compared with healthy controls. Wound scratch assay and Transwell method manifested that TIPE2 can down-regulate the invasion and metastasis of hepatoma cell. These results suggested that TIPE2 as a novel tumor suppressor may plays an important biological role in the development and progression of hepatocellular carcinoma, thus may be a promising strategy for diagnosis and targeted treatment of HCC.
