Galectin-3在哮喘小鼠气道炎症中的作用及低分子柑桔果胶的干预  被引量:1

摘  要:目的探讨小鼠肺组织半乳糖凝集-3(Galectin-3)表达水平及其与支气管哮喘的关系,研究低分子柑桔果胶(LCP)对哮喘小鼠肺组织Galectin-3表达及TH1/TH2平衡的影响。方法用卵清白蛋白建立哮喘模型。将BALB/c小鼠随机分为五组:正常对照组(A组)、哮喘组(B组)、低分子柑桔果胶预防组(C组)、低分子柑桔果胶治疗组(D组)和地塞米松治疗组(E组),每组10只。留取支气管肺泡灌洗液(BALF)进行细胞总数计数和分类计数;应用双抗体夹心酶联免疫吸附试验方法检测BALF中IL-4、IFN-γ浓度,采用免疫组化和RT-PCR法分别检测Galectin-3蛋白和Galectin-3验mRNA的表达。结果哮喘组肺组织中Galectin-3蛋白和Galectin-3 mRNA表达的强度均高于对照组,LCP预防组和治疗组均显著低于哮喘纽,差异有统计学意义(P〈0.05,P〈0.01)。Galectin-3及其mRNA表达与BLAF中的IL-4浓度成显著正相关(r=0.791、0.761,P〈0.01),而与BLAF中的IFN-γ浓度成显著负相关(P=0.668、0.634,P〈0.01)。结论哮喘小鼠肺组织Galectin-3高度表达,参与气道炎症的发生,LCP干预后下调Galectin-3表达,纠正Th1/Th2失衡。Objective To evaluate the expression of galectin-3 in mice lung tissue, and to explore the role of galectin-3 in the bronchus of a mouse model of asthma. To access the effect of Low-Molecular-Weight Citrus Pectin ( LCP ) on the expression of Galectin-3, Galectin-3mRNA and the level of Th1/Th2 balance. Methods Fifty male BALB/c mice were randomly divided into five groups : the control group, asthma group, LCP prevented group and LCP treated group and dexamethasone treated group.In the experiment, the mouse model of asthma was established by the ovalbumin ( OVA ) challenge Methods. Counts of total cell numbers and differentiated cell numbers in bronchoalveolar lavage .The protein expression of Galectin-3 was detected by Immuno-histochemistry ( IHC ) ; the mRNA expression of Galectin-3 was detected by transcriptase polymerase chain reaction ( RT-PCR ) . Results Immunohistochemical staining and transcriptase polymerase chain reaction showed that the protein content of Galectin-3 and Galectin-3mRNA expression in the lung tissue of asthma group were significantaly higher than that of the control group, while those of LCP prevented group and LCP treated group were significantly lowed than asthma group. ( P〈0.05 or P〈0.01 ) . The Gatectin-3 and the Galectin-3mRNA expression had positive correlation with the concentration of IL-4 in BALE ( r=0.791, 0.761, P〈0.01 ) while had negative correlation with the IFN-γ in BALE ( r=-0.668, 0.634, P〈0.01 ) Conclusion Galectin-3mRNA and Galectin-3 were strongly expressed in airway and lung of sensitized mouse. LCP candownregulate expression of Galectin-3mRNA, reduce airway inflammation and modulate Th1/Th2 balance.
