摘 要:目的研究新疆地区维吾尔族人群TCF7L2基因单核苷酸多态性与2型糖尿病的相关性。方法 2012年3月—2013年3月,应用基质辅助激光解吸附电离飞行时间质谱分析质谱技术(MALDI-TOF),对新疆医科大学第一附属医院就诊的维吾尔族2型糖尿病患者969例(T2DM组)和958例体检健康者(NC组)的rs12255372和rs7085532进行基因分型;同时进行人体测量学及代谢指标的检测。结果两组rs12255372多态性位点3种基因型,T、G等位基因频率比较,差异均有统计学意义(P<0.05)。非条件Logistic回归分析显示,T等位基因携带者患2型糖尿病的风险为G等位基因携带者的1.238倍〔OR=1.238,95%CI(1.049,1.461),P<0.05〕;校正年龄、性别后GT基因型携带者患2型糖尿病的风险为GG基因型携带者的1.221倍〔OR=1.221,95%CI(1.001,1.490),P<0.05〕;GT+TT基因型携带者患2型糖尿病的风险为GG基因型携带者的1.252倍〔OR=1.252,95%CI(1.033,1.516),P<0.05〕。两组rs7085532多态性位点3种基因型,A、G等位基因频率比较,差异均无统计学意义(P>0.05)。结论 TCF7L2基因rs12255372多态性位点可能与新疆维吾尔族人群2型糖尿病相关,而rs7085532多态性位点可能与新疆维吾尔族人群2型糖尿病无相关性。Objective To study the association between transcription factor 7 - like 2(TCF7L2)polymorphisms and type 2 diabetes mellitus( T2DM ) in Uygur nationality in Xinjiang. Methods From March 2012 to March 2013,2 polymorphisms( rs12255372 and rs7085532)of TCF7L2 gene were genotyped in 969 T2DM patients from Xinjiang Medical University,the First Affiliated Hospital of Uygur(T2DM group)and 958 normal subjects(NC group)by matrix - assisted laser desorption/ ionization - time of flight( MALDI - TOF). At the same time,the anthropometry and metabolism indices were measured. Results T allele frequency and GT,TT,3 genotypes frequencies of rs12255372 were significantly different between T2DM group and NC group(P 〈 0. 05). The risk of T2DM was 1. 238 times higher in patients with T allele than in those with G allele〔OR = 1. 238,95% CI(1. 049,1. 461),P 〈 0. 05〕,1. 221 times higher in patients with GT genotype than in those with GG genotype after adjusting age,gender〔OR = 1. 221,95% CI(1. 001,1. 490),P 〈 0. 05〕,1. 252 times higher in patients with GT + TT genotype than in those with GG genotype〔OR = 1. 252,95% CI(1. 033,1. 516),P 〈 0. 05〕. There was no significant difference in 3 genotypes,A,G alleles of rs7085532 polymorphic sites between the 2 groups( P 〉 0. 05). Conclusion The rs12255372 polymorphisms in TCF7L2 gene may be associated with T2DM,while rs7085532 polymorphisms may not in Uygur population in Xinjiang region.
