摘 要:目的比较乙酰天麻素和天麻素灌胃给药对糖尿病大鼠视网膜神经节细胞(retinal ganglion cells,RGC)的保护作用。方法取56只SD大鼠随机分为7组:对照组、糖尿病模型组、生理盐水治疗组、乙酰天麻素低浓度组及高浓度组、天麻素低浓度组及高浓度组,每组8只。链脲佐菌素腹腔注射建立SD大鼠糖尿病模型,血糖高于16.7 mmol·L-1确定为造模成功。模型鼠出现高血糖1周后,分别给予100 mg·kg-1、200 mg·kg-1乙酰天麻素及50 mg·kg-1、100 mg·kg-1天麻素溶液灌胃给药8周,对照组只给予生理盐水灌胃。模型组和对照组大鼠处死后摘除眼球,做视网膜冰冻切片,PI染核,观察神经节细胞层细胞数量变化并计数。结果造模后8周,糖尿病模型组和对照组的细胞计数分别为5.50±4.50和24.75±3.50,差异有统计学意义(P<0.05)。糖尿病大鼠给予天麻素灌胃给药后,神经节细胞层的RGC细胞的损伤减轻。免疫荧光显示低、高浓度组糖尿病大鼠RGC细胞数量较糖尿病模型组大鼠及对照组大鼠增加,其中高浓度天麻素组增加更明显,生理盐水治疗组及低、高浓度天麻素组的细胞计数分别为6.10±3.75、14.00±3.50和18.01±4.00,差异有统计学意义(P<0.05)。糖尿病大鼠给予乙酰天麻素治疗后,神经节细胞层的RGC细胞数量较糖尿病模型组增加,其中高浓度组增加更明显,低、高浓度乙酰天麻素组的细胞计数分别为18.25±5.50和21.88±5.50,差异有统计学意义(P<0.05);与天麻素组比较,高浓度乙酰天麻素组RGC的细胞数量明显增加,差异有统计学意义(P<0.05)。结论乙酰天麻素比天麻素能更有效保护糖尿病RGC。Objective To compare the protective effects of acetagastrodin and gastrodin by intragastric administration on retinal ganglion cells (RGC) in diabetic model. Methods Fifty-six SD rats were divided into seven groups as follow: Control group ,diabetes model group ,physiological saline group ,low and high concentration acetagastrodin group,low and high concentration gastrodin group. SD rat diabetic models were established by streptozotocin intraperitoneal injection, which was considered as success when the blood sugar was measured over 15.7 mmol ·L^-1. The model rats were given 100 mg·kg^-1 ,200 mg·kg^-1 acetagastrodin and 50 mg ·kg^-1 ,100 mg·kg^-1 gastrodin respectively by intragastric administration for 8 weeks. Physiological saline group only received physiological saline by intragastric administration for 8 weeks. All rats eyeballs were executed and made into frozen sections. The nuclear was stained by PI. RGC in ganglion cell layer was observed and counted. Results The number of ceils in ganglion cell layer in diabetic rats (5.50 ±4.50) decreased compared with normal rats (24.75 ±3.50), there was statistical difference (P 〈 0.05 ). Ceil count in acetagastrodin group and gastrodin group was increased obviously compared with physiological saline group, which showed concentration dependent. Cell count in physiological sa- line group,low and high concentration gastrodin group were 6.10 ±3.75,14.00 ±3.50 and 18.01 ±4.00 ,respectively,there was statistical difference (P 〈0.05). Cell count in low and high concentration acetagastrodin group were 18.25 ±5.50 and 21.88 ±5.50, respectively, there was statistical difference (P 〈 0.05 ). Cell quantity in high concentration acetagastrodin group was higher than that in high concentration gastrodin group,there was statistical difference (P 〈 0. 05 ). Conclusion Acetagastrodin has more protective effects on RGC against apoptosis than gastrodin.
