摘　　要：目的:探讨claudin-4(CLDN4)在乳腺癌中的表达及临床意义。方法:收集本院外科手术切除的60例乳腺癌及对应癌旁乳腺组织,实时定量PCR(qRT-PCR)检测CLDN4mRNA在乳腺癌及癌旁组织中的表达;免疫组织化学染色检测CLDN4表达,统计分析CLDN4蛋白与肿瘤临床病理资料间的相关性。结果:CLDN4mRNA及蛋白在乳腺癌组织中表达水平明显高于对应癌旁组织,差异具有统计学意义(P<0.05);乳腺癌组织中CLDN4蛋白高表达与肿瘤体积增大、恶性组织学分级、淋巴结转移及高TNM分期成显著正相关(P<0.05)。结论:CLDN4在乳腺癌组织中表达上调并与肿瘤恶性临床病理特征有关。CLDN4可能成为乳腺癌早期诊断的重要标志物及分子治疗靶点之一。Objective： To investigate the expression and clinical significance of claudin-4 in human breast cancer. Meth- ods： Real-time quantitative PCR （qRT-PCR） was applied to detect the mRNA expression of claudin-4 in breast cancer（n= 60） in comparison to the corresponding tumor-adjacent normal tissues. The protein expression of claudin-4 was measured by immu- nohistochemistry （IHC）. Results： The mRNA and protein expression level of claudin-4 in breast cancer tissues was significant- ly higher than in adjacent normal tissues （P〈0.05）. The positive expression of claudin-4 protein was also associated with large tumor size, high histological grading, lymphatic metastasis, and advanced TNM stage（P〈0.05）. Conclusions： The expression of claudin-4 in breast cancer tissues was significantly higher than in tumor-adjacent normal tissues, and associated with tumor malignant clinicopathological features claudin-4 may become a new diagnosed mraker and effective therapeutic target.
