摘  要:目的:比较两种方法建立阿尔茨海默病转基因小鼠模型对小鼠脑新皮质区Aβ分布的影响。方法:选取16月龄雄性小鼠20只,分别建立APP/PSl双转基因(2×Tg-AD组)模型与APP/PSl/tau三转基因(3×Tg-AD组)模型。取两组小鼠新皮质区脑组织行6E10单克隆抗体免疫组化染色等方法显示Aβ阳性神经元及斑块,观察其分布与形态差异,并利用图像分析系统定量比较。结果:在新皮质区2×Tg-AD组Aβ阳性产物主要位于细胞外即细胞外Aβ(eAβ),形成大量的老年斑,细胞内阳性产物少;而3×Tg-AD组Aβ阳性产物主要位于神经元细胞内即细胞内Aβ(iAβ),但老年斑少见。结论:2×TgAD组与3×Tg-AD组Aβ阳性产物在新皮质区分布的差异可能反映了两种AD小鼠模型神经病理等改变的不同。Objective: To compare effect of different transgenic mice model with Alzheimer's disease (AD) on distribu- tion of amyloid beta-protein in neocortex. Methods: A total of 20 mice with 18-month old were selected, APP/PSI double transgenic mice model (2× Tg-AD) and APP/PS1/tau triple transgenic mice model (3 × Tg-AD) were established in this exper- iment. The brain tissues in neocortex were extracted, and 6E10 immunohistochemical staining was applied to observe the distribution and morphological features of Aβ positive products. Image analysis system was applied to quantitatively analyze the difference of the products. Results: The deposition of Aβ positive products in the neocortex of 2 × Tg-AD was mainly extracel- lular Aβ positive products (eAβ), forming a lot 05 positive plaques, with few intracellular positive product. While the positive products were distributed as intracellularly Aβ positive products (iAβ) in 3 × Tg-AD group, but the Aβ positive plaques were rarely seen. Conclusion: The differences in distribution of Aβ positive products in the neocortex between the 2 X Tg-AD and 3 × Tg-AD mice likely suggest the differences in neuropathological changes of the two AD models.
