肝癌患者血清Th1/Th2细胞因子含量检测及其与机体免疫功能的关系

摘　　要：目的:研究肝癌患者血清中Th1/Th2细胞因子的含量及其与机体免疫功能的关系。方法:将在本院就诊的肝癌患者纳入研究的观察组,同期在我院体检的健康者纳入研究的对照组,采用酶联免疫吸附法检测两组受试者血清中Th1、Th2细胞因子以及补体C3和C4的含量,流式细胞法检测T细胞亚群含量。结果:(1)Th1/Th2细胞因子:观察组的Th1细胞因子肿瘤坏死因子-a(TNF-a)、干扰素-γ(IFN-γ)含量低于对照组,Th2细胞因子白介素-6、10(IL-6、IL-10)含量高于对照组(P<0.05);(2)免疫功能:观察组的CD4+T细胞、CD16/65+NK细胞、补体C3和C4含量低于对照组,CD8+T细胞的含量高于对照组(P<0.05);(3)细胞因子与免疫功能的关系:TNF-a、IFN-γ含量与CD4+T细胞、CD16/65+NK细胞、补体C3和C4的含量呈正相关,与CD8+T细胞含量呈负相关;IL-6、IL-10含量与CD4+T细胞、CD16/65+NK细胞、补体C3和C4的含量呈负相关,与CD8+T细胞含量呈正相关。结论:肝癌患者存在Th1/Th2漂移,血清中Th1细胞因子TNF-a和IFN-γ含量降低、而Th2细胞因子IL-6、IL-10含量升高,且与机体的天然免疫和细胞免疫功能密切相关。Objective： To study the relationship of serum Th1/Th2 cytokines with immune function of patients with hepatocellular carcinoma. Methods： Patients with hepatocellular carcinoma in our hospital from 2012 January to 2014 March were enrolled in observation group and healthy subjects in the same period were enrolled in control group. Then serum Th1, Th2 cytokines and complement C3 and C4 content were assayed by ELISA; T cell subsets contents were determined by flow cytometry assay. Results： Th1 cytokines TNF-α, IFN-γ content of observation group were significantly lower than control group, Th2 cytokines IL-6, IL 10 content were significantly higher than control group（P〈0.05）. CD4＋ T cells, CD16/65 NK cells, complement C3 and C4 content of observation group were significantly lower than control group（P〈0.05）, CD8 ＋ T ceils content in observation group was significantly higher than control group（P〈0.05）. TNF-α, IFN-γcontents were posi- tively correlated with CD4＋ T cells, CD16/65＋ NK cells, complement C3 and C4 content and negatively correlated with CD8＋ T cells content; IL-6, IL-10 content were negatively correlated with CD4＋ T cells, CD16/65＋ NK ceils, complement C3 andC4 content and positively correlated with CD8＋ T cells content. Conclusion： Patients with hepatocellular carcinoma have Th1/ Th2 drift, decreased Th1 cytokines TNF-α, IFN-γ contents and increased Th2 cytokines IL-6, IL-10 contents. They are closely related to natural immunity and cell immunity.
