摘 要:目的探讨miRNA-143对前列腺癌细胞增殖的影响及其作用机制。方法通过RT-PCR和Western blot方法检测miRNA-143及其靶基因KRAS在前列腺癌组织中的表达;用MTT方法和集落形成实验研究miRNA-143对激素非依赖性前列腺癌DU145细胞增殖的影响。结果 miRNA-143及其靶基因KRAS在前列腺癌组织中的表达呈负相关;miRNA-143高表达对DU145细胞的增殖具有抑制作用;DU145细胞中miRNA-143高表达抑制KRAS-ERK信号通路的激活及Cyclin D1的表达。结论 miRNA-143通过RAS/MAPK信号通路抑制前列腺癌的增殖,为激素非依赖性前列腺癌治疗提供了分子生物学基础。Objective To verify its effect and mechanisms on prostate cancer cell proliferation. Methods miR.NA-143 and its target gene KRAS expression was measured by real-time PCR and western blotting and effects of miRNA-143 in cell proliferation and migration were evaluated by MTT and colony formation. Results An inverse correlation between miRNA-143 and KRAS expression in prostate cancer tissues. Over-expression of miRNA-143 in DU145 cells suppressed their proliferation by targeting RAS/MAPK pathway. Conclusion miRNA-143 plays an important role in prostate cancer cell proliferation by suppressing KRAS and subsequent inactivation of MAPK pathway, which provide a new approach for HRPC treatment.
