健脾复方对大肠癌CD44和β-catenin表达的调控作用

摘　　要：目的探讨健脾复方对大肠癌SW620细胞株CD44和β-catenin表达的调控作用。方法采用体外实验,将SW620细胞分别与正常培养基(空白对照组)、正常裸鼠血清培养基(药物空白组)和含药血清培养基(健脾复方组)培养72 h后,采用流式细胞技术检测CD44的表达情况,Western Blot方法检测β-catenin的表达情况。结果空白对照组、药物空白组和健脾复方组的CD44相对表达量分别为0.791±0.008,0.737±0.007,0.533±0.018。经方差分析,3组间比较差异有统计学意义(P<0.001);经LSD法两两比较,健脾复方组与空白对照组、健脾复方组与药物空白组比较差异均有统计学意义(P均<0.05)。空白对照组、药物空白组和健脾复方组的β-catenin相对表达量分别为1.396±0.437,1.491±0.467,0.671±0.213。经方差分析,3组间比较差异有统计学意义(P=0.004);经LSD法两两比较,健脾复方组与空白对照组、健脾复方组与药物空白组间比较差异均有统计学意义(P均<0.05)。结论健脾复方能抑制透明质酸与CD44的结合以及β-catenin的表达,从而起到抗肿瘤的作用。Objective It is to investigate the regulation of Jianpi Compound Recipe on expression of CD44 and β -catenin in colorectal cancer cell line SW620. Methods An in vitro study was adopted. SW620 cell was cultured in normal culture medium （blank control group） , serum medium （drug blank group） , and Jianpi Compound Recipe contained serum medium （TCM group）. After 72-hour of cultivation, the expression of CD44 was detected by flow cytometer, and β -catenin was de- tected by Western Blot. Results The expression of CD44 in blank control group, drug blank group, and TCM group was 0.791 ± 0. 008, 0. 737 ± 0. 007, and 0. 533 ± 0. 018 respectively. ANOVA analysis showed that there was significant difference among three groups （P 〈 0. 001 ） , and further LSD analysis showed that there was significant difference between TCM group and blank control group or drug blank group respectively （ all P 〈 0.05 ）. The expression of β- catenin in blank control group, drug blank group, and TCM group was 1. 396 ± 0. 437, 1. 491 ± 0. 467, and 0. 671 ± 0. 213 respectively. ANOVA analysis showed that there was significant difference among them （P = 0. 004） , and further LSD analysis showed there was significant difference between TCM group and blank control group or drug blank group respectively （ all P 〈 0.05 ）. Conclusion Jianpi Compound Recipe can regulate the expression of CD44 and β - catenin to achieve its anti-cancer role.
