摘 要:目的制备磁性隐形阿霉素脂质体,对其理化性质及其热疗后对SKOV-3细胞的影响进行评价。方法使用pH梯度法制备磁性隐形阿霉素脂质体,通过透射电镜观察其形态,通过紫外分光光度计测试其包封率,差示扫描量热仪测定其相变温度,MTT及流式细胞仪测定凋亡率。结果不同脂质体粒径在80-150nm间,包封率达到76%,相变温度为42.3℃,MTT及流式的结果均显示热疗对SKOV-3细胞的杀伤作用显著高于对照组。结论磁性隐形阿霉素脂质体包封率较高,粒径较小,对SKOV-3具有显著影响。Objective Aim To prepare the doxorubicin-loaded Fe3 O4 magnetic nanoparticles modified with PEG,to study the physicochemical properties of the nanoparticles and the effects on SKOV-3 cells after hyperthermia.Methods Using the pH gradient method to prepare the liposomes,the characteristics were observed with TEM,UV spectro-photometer,DSC,and the apoptosis rate was determined by MTT and flow cytometry.Results The particle size of li-posomes was during 80-150 nm,the encapsulation ratio detected by UV spectrophotometer spherical was 76%.The DSC showed the liposomes changed their phase at 42.3℃.The result of MTT and flow cytometry showed cytotoxic effect of hyperthermia on SKOV-3 cells was significantly better than that of control group.Conclusion These doxoru-bicin liposomes with high encapsulation ratio and small particle size might be high efficiency on SKOV-3 cells.
