青蒿琥酯诱导人前列腺癌细胞凋亡的实验研究

摘　　要：目的:探讨青蒿琥酯(artesunate,Art)对人前列腺癌PC-3细胞株抑制作用及机制。方法:进行前列腺癌PC-3细胞培养,不同浓度Art干预,采用MTT法检测用药后细胞增殖率的改变,流式细胞术(FCM)检测细胞周期的改变和凋亡比。结果:与对照组比较,Art可显著抑制PC-3细胞增殖(P<0.01);PC-3细胞主要被阻滞在G0/G1期,FCM检测PC-3细胞凋亡比,各实验组均明显高于对照组(P<0.01),且呈时间、剂量依赖性。结论:Art可抑制PC-3细胞增殖,其作用机制可能与调控细胞周期、诱导细胞凋亡有关。Objective ：To investigate the action of artesunate on the inhibition of PC - 3 cell of prostate cancer, and its mechanism. Methods ： PC - 3 cell was cultured with different concentration artesunate, the proliferation of PC - 3 were examined by MTr, the cell ratio in the different stages of the cell cycle and apoptosis ratio of PC - 3 were exam- ined by flow cytometry（FCM）. Results：Compared with the control group,the proliferation of PC -3 was significantly inhibited by Art（P 〈0.01 ）. The cell cycle of PC-3 was mainly blocked at G0/GI stage;compared with the control group,the apoptosis percentage of the PC -3 in every experiment groups were higher （P 〈 0.01 ） , and Art could in- hibit the proliferation and induce apoptosis of PC - 3 cell in a time - dose dependent manner. Conclusion ： Art could inhibit the proliferation of PC -3 cell, and mechanism is related with modulating cell cycle distribution and inducing PC - 3 apoptosis
