摘　　要：目的:探讨mi R-451对鼻咽癌细胞增殖、侵袭和转移能力的影响。方法:选取正常鼻咽上皮细胞株NP69及4株鼻咽癌细胞株CNE-1、CNE-2、5-8F、6-10B,荧光定量PCR检测mi R-451的表达量。选取鼻咽癌细胞株CNE-1和CNE-2,分别转染mi R-451 mimics和阴性对照后,采用MTT法检测鼻咽癌细胞的增殖能力,划痕实验检测其侵袭能力,transwell迁移实验检测其迁移能力。通过生物信息学方法预测mi R-451的靶基因。结果:与正常鼻咽上皮细胞相比,mi R-451在鼻咽癌细胞株中的表达量明显降低。鼻咽癌细胞过表达mi R-451后,其增殖、侵袭和迁移能力均受到抑制。通过靶基因预测数据库预测RAB14为mi R-451的靶基因,并通过荧光素酶报告基因实验、RT-PCR、Western blot实验验证了这一结果。将RAB14低表达后,也在一定程度上抑制了鼻咽癌的增殖、侵袭和转移能力。结论:mi R-451通过靶向RAB14抑制了鼻咽癌细胞的增殖、侵袭和迁移能力。Objective To investigate the effect of miR-451 on the proliferation, invasion and migration of nasopharyngeal carcinoma （NPC） ceils. Methods Quantitative reverse transeription-polymerase chain reaction （qRT-PCR） was used to detect miR-451 expression in normal human nasopharyngeal epithelial cell NP69 and another four NPC cell lines. MTT assay, wound scratch assay and transwell migration assay were performed to study the role of miR-451 in NPC proliferation, invasion and migration, respectively. Identifieationa of target gene of miR-451 was confirmed by luciferase reporter assays, real-time PCR, and western blot assay, respec- tively. Results Expression of miR-451 was lower in NPC cell lines than in the normal nasopharyngeal epithelial cell. MiR-451 was shown to inhibit the proliferation, invasion and migration of NPC cells. Additionally, we found that miR-451 directly targets RAB14 （ras-related protein 14）. Knockdown of RAB14 could inhibit the pro- liferation, invation and migration of NPC cells as well. Conclusions MiR-451 inhibits the proliferation, invasion and migration of nasopharyngeal carcinoma cells through targeting RAB14.
