摘 要:目的观察大豆异黄酮(SIF)对动脉硬化小鼠胆固醇外排作用的影响,揭示SIF降脂及抗动脉硬化的作用靶标和相关机制,为进一步探索(SIF)的功能和动脉硬化的防治新措施提供依据。方法 C57BL/6J小鼠30只,随机分为3组,每组10只。正常对照组饲以正常合成饲料;模型对照组和大豆异黄酮组均饲以高脂饲料,后者再以大豆异黄酮灌胃,实验周期16周。观察大豆异黄酮干预对小鼠主动脉形态结构、三磷酸腺苷结合盒转运体A1(ABCA1)表达和血清胆固醇水平的影响。结果与模型对照组比较,大豆异黄酮组小鼠主动脉内皮未出现明显增厚,脂质沉积和泡沫细胞也明显减少;主动脉ABCA1蛋白表达显著增强(P<0.05),接近正常对照组水平;血清总胆固醇和低密度脂蛋白胆固醇分别为(2.31±0.19)和(1.65±0.19)mmol/L,明显低于模型对照组水平〔(2.59±0.22)和(2.12±0.14)mmol/L,P<0.05〕;血清高密度脂蛋白胆固醇为(1.19±0.16)mmol/L,明显高于模型对照组〔(0.74±0.19)mmol/L,P<0.01〕。结论大豆异黄酮通过诱导ABCA1表达,增强组织胆固醇的逆转运,改善脂质代谢状况,从而发挥抗动脉粥样硬化的作用。Objective To study the effect of soy isoflavone on cholesterol efflux in atherosclerosis mice, to reveal the targets and mechanisms of soy isoflavone ameliorating lipid metabolism and antiatherogenic effect, so as to provide the basis for further study on its function, and new measures to prevent and control atherosclerosis. Methods The atherosclerosis model was developed by feeding C57BL/6J mice a high-fat diet for sixteen weeks. The morphologic change of aortic walls was examined, and the effects of soy isoflavone on ATP-binding cassette transporter A1 (ABCA1) protein expression and blood cholesterol were observed. Results Compared with model control group, the pathologic changes in aortic wall (endotheliun thickening, lipid deposition, and foam cells ap- pearance) in soy isoflavone group were obviously improved. The expression of aorta ABCA1 in soy isoflavone group was raised and approached normal control level. Serum total cholesterol and LDLC content in soy isoflavone group (2.31±0.19), ( 1.65±0. 19) mmol/L respectively] were significantly lower than in model control group[ (2.59±0.22), (2.12±0.14) mmol/L, P〈0.053, and higher than in normal control group] (2.11 ±0. 12 ), ( 1.10±0.11 ) mmol/L,P〈0.05]. Moreover,serum HDLC content in soy isoflavone group [ (1.19±0.16) mmol/L] was signifi- eontly higher than in model control group [ (0.74±0. 19 ) retool/L, P〈0.01]. Conclusion Soy isoflavone could strengthen reverse cholesterol transport and improve lipid metabolism by inducing ABCA1 expression, thus to bring out its antiatherosclerotic effect.
