摘　　要：目的 建立人胃癌细胞株HGC-27裸小鼠皮下移植瘤模型,为胃癌的研究提供简单实用的载体。方法 培养HGC-27人胃癌细胞,达106~107个活细胞/瓶,以0.9%氯化钠无菌水溶液为溶剂制成单细胞悬液,随机注射到裸小鼠左腋下或右腋下,观察肿块的形成,并进行组织学观察。结果 注射后8 d触及米粒般大小的肿块,14 d可见肿块明显隆起。组织学观察癌细胞无腺体样结构形成,呈片状、巢状或条索状排列,细胞形状不规则,呈圆形、梭形或不规则形,核大深染,核质比增高,可见核分裂象。结论 人胃癌细胞株HGC-27裸小鼠皮下移植瘤模型的建立,操作简便,肿块表浅,可直接观测肿瘤,为进一步的研究能提供简单有效的模型。Objective To establish a subcutaneous tumor xenograft model in nude mice by injection of human gastric carcinoma HGC - 27 cells and to provide references to related researches. Methods HGC - 27 cell line was cultured and made into single - cell suspension of 10^6- 10^7 in normal saline . HGC - 27 cells were injected subcutaneously into nude mice. The oncogenesis was observed and the organization structure was observed through light microscope. Results The tumors were not observed till the 8th day of injection, but solid granule - like mass could be felt. The oncogenesis was evident at, the 2nd week. Under the light microscope, the morphol- ogy of HGC - 27 transplanted tumors show the characteristic of cancer. Conclusion The transplanted tumor model of HGC -27 cell in nude mice is established successfully. It provides a foundation for the experimental studies of gastric carcinoma.
