CDl57在胃癌组织中的表达与临床意义及其对胃癌细胞侵袭的影响

摘　　要：目的探讨CDl57蛋白在人胃癌组织中的表达情况及其与胃癌患者临床病理指标和预后的关系；初步探讨CDl57对胃癌AGS细胞侵袭转移的作用及其机制。方法用免疫组织化学方法检测CDl57蛋白在90例胃癌及配对癌旁组织中的表达，分析其表达结果与胃癌患者的临床病理资料和预后的相关性。通过小干扰RNA（small interfering RNA，siRNA）转染人胃癌AGS细胞下调CDl57的表达，用Transwell侵袭实验检测CDl57对胃癌AGS细胞侵袭能力的影响。结果CDl57蛋白在胃癌组织及癌旁组织的表达率分别为67％和12％，差异有统计学意义（X^2=55．84，P〈0．01）；CDl57蛋白的表达水平与肿瘤浸润深度（X^2=12．503，P〈0．01）、淋巴结转移（X^2=8．693，P=0．003）和远处转移（X^2=4．944，P=0．027）相关，而与年龄（X^2=1．659，P=0．198）、性别（X^2=1．431，P=0．232）和分化程度（X^2=0．407，P=0．856）无相关；CDl57高表达组胃癌患者中位生存期短于低表达组（29．2个月比46．0个月，X^2=4．438，P=0．036）。与对照组相比，siRNA组CDl57RNA表达（t=45．004，P〈0．01）和蛋白表达（t=32．877，P〈0．01）降低，侵袭能力减弱（F=98．455，P〈0．01）。结论CDl57高表达与胃癌的浸润、转移和不良预后有关，下调CDl57表达可抑制胃癌细胞侵袭能力。Objective To investigate the expression of CD157 in gastric cancer and its relationship with clinicopathological parameters and prognosis, and to explore the effects and molecular mechanisms of CD157 in invasion and metastasis of gastric cancer cells. Method The expression of CD157 in 90 cases of gastric cancer tissues and corresponding adjacent non-cancerous tissues （ANCT） was detected using immunohistochemistry. The correlation between the expression of CD157 and elinicopathologieal parameters and prognosis was analyzed statistically. Human gastric cancer AGS cells in vitro were transfeeted with small interfering RNA to down regulate the expression of CD157. The invasion capability of gastric cancer cells were assessed by Transwell assays. Result High expression of CD157 protein was observed in 67% gastric adenocarcinoma compared with that in 12% ANCT （ X^2 = 55.84, P 〈 0.01 ）. The expression of CD157 was correlated with the depth of tumor infiltration （X^2 = 12. 503, P 〈 0. 01 ）, lymph node metastasis （ X^2 = 8. 693, P = 0. 003） and distant metastasis （ X^2 = 8. 693, P = 0. 003）, but not with age （ X^2 = 1. 659, P = 0. 198）, gender （ X^2 = 1.431, P = 0. 232） or tumor differentiation （ X^2 = 0. 407, P = 0. 856）. Patients with high expression of CD157 had shorter survival （29.0 ± 16. 7 months） than those with low expression（46.0 ± 18.2 months） （X^2 = 4. 438,P = 0. 036）. AGS cells interfered by siRNA has lower expression of CD157 both in protein （t = 32. 877, P 〈 0.01 ） and RNA （ t = 45. 004, P 〈 0. 01 ） levels. Down regulated expression of CD157 could inhibit invasion potential of gastric cancer cells （F = 98.455,P 〈 0.01 ）. Conclusions High expression of CD157 might play an important role in the poor survival of gastric cancer. Down-regulation of CD157 can reduce invasive ability of gastric cancer cells.
