摘 要:目的探讨土荆皮酸对肺癌细胞系A549细胞增殖和凋亡的影响及其可能的作用机制。方法体外培养的A549细胞,用不同浓度的土荆皮酸(0、4、8、16、32μmol/L)处理24、48、72 h,MTT法测定其对A549细胞增殖的影响;土荆皮酸(0、4、8、16μmol/L)处理A549细胞48 h后,流式细胞仪检测细胞凋亡和周期,分光光度计检测Casepase-3相对活性,Western blot法检测PTEN、Akt、p-Akt的表达。结果土荆皮酸能有效抑制A549细胞增殖,且呈浓度和时间依赖性(P<0.05);土荆皮酸作用A549细胞48 h后,细胞凋亡率从14.8%上升至37.7%,显著高于对照组(土荆皮酸0μmol/L)(P<0.05);流式细胞仪检测结果显示,土荆皮酸能抑制细胞的G1期行进和G1/S转换;Casepase-3相对活性显著增加(P<0.05);土荆皮酸可上调PTEN表达并抑制p-Akt的表达(P<0.05)。结论土荆皮酸可抑制A549细胞增殖并促进其凋亡,其作用机制可能与上调PTEN并抑制Akt信号通路,继而阻滞细胞周期并激活线粒体凋亡途径相关。Objective To investigate the effects of Pseudolaric acid B on the cell proliferation and apoptosis in lung cancer line A549 cells and discuss the possible mechanisms. Methods A549 cells were cultured in vitro. After treatment by Pseudolaric acid B of different concentrations(0, 4, 8, 16, 32 μmol/L) respectively for 24, 48, 72 h, the influence of Pseudolaric acid B on the proliferation of A549 cells were determined by the MTT method. After treatment by Pseudolaric acid B of different concentrations(0, 4, 8, 16 μmol/L) for 48 h, the cells apoptosis and cycle were detected by flow cytometry; the relative activity of caspase-3 was monitored by spectrophotometer; the expression of PTEN, Akt and p-Aktproteins were detected by the Western blot. Results From the data of MTT, the cell proliferation of A549 cells was inhibited by Pseudolaric acid B, and in a dose-dependent and time-dependent manner(P 〈0.05). Annexin V-FITC/PI staining showed that Pseudolaric acid B significantly induced cell apoptosis. After treated with Pseudolaric acid B, the apoptosis rate of A549 cells increased from 11.6% to 34.5%, which showed an obvious concentration-effect relationship(P 〈0.05). Flow cytometry assays demonstrated that Pseudolaric acid B blocked the cell cycle at the G1 and G1to S transition, and the percentage of S-phase cells decreased obviously. The relative activity of caspase-3 of Pseudolaric acid B was increased(P 〈0.05). The treatment with Pseudolaric acid B promoted the expression of PTEN,and suppressed the expression of p-Aktin a dose-dependent manner(P 〈0.05). Conclusion Pseudolaric acid B inhibits the cell proliferation and induces cell apoptosis in A549 cells, and the effects of antitumor may be associated with up-regulation of PTEN and inhibition of the Akt signaling pathway, thus blocking the cell cycle and activating mitochondria apoptosis pathway.
