摘  要:[目的]观察中国汉族和朝鲜族8个Y-STR基因座遗传多态性在法医学个人识别和亲权鉴定中的适用性.[方法]利用PCR扩增、变性聚丙烯酰胺凝胶电泳和银染方法对各200名中国汉族和朝鲜族人群的DYS441~DYS448 8个Y-STR基因座单倍型进行检测和分析.[结果]在汉族群体中分别检测出7,5,6,7,4,10,10和6个等位基因,基因多样性分布为0.540 1~0.819 0,8个基因座联合单倍型为189种,基因多样性为0.999 2;在朝鲜族群体中分别检测出8,6,6,5,6,10,9和6个等位基因,基因多样性分布为0.513 5~0.799 6,8个基因座联合单倍型为174种,基因多样性为0.997 9.[结论]8个YSTR基因座在2个群体中均具有较高的遗传多样性,是较理想的遗传标记系统,对中国汉族和朝鲜族的个人识别及亲权鉴定等具有较高的应用性.OBJECTIVE To observe the genetic polymorphism at 8 Y-STR loci in Han and Korean-Chinese nationalities to application in personal and paternity identification.METHODS Using the PCR,polyacrylamide gel electrophoresis and silver staining,the haplotype polymorphisms of DYS441-DYS448Y-STR loci were detected and analyzed in 200 unrelated Han and Korean-Chinese nationalities individuals.RESULTS7,5,6,7,4,10,10 and 6alleles were respectively detected in Han nationality,and the gene diversity was distributed in 0.540 1-0.819 0,and the combind haplotype of 8 Y-STR loci was189 types.8,6,6,5,6,10,9 and 6 alleles were respectively detected in Korean-Chinese nationality,and the gene diversity was distributed in 0.513 5-0.799 6,and the combind haplotype of 8 Y-STR loci was174 types.CONCLUSIONThe 8 Y-STR loci have high gene diversity in two population as useful genetic markers to application in personal and paternity identification between Han and Korean-Chinese nationalities.
